Hippophae rhamnoides(Hippophae rhamnoides var.sp) is deciduous shrubs or small trees of sea-buckthorn plant of Elaeagnaceae and cultivation of a larger area in the country. Seabuckthom oil rich of variety substances,is the highest value-added industries Sea-buckthorn material.In order to improve the utilization value and economic value of Hippophae rhamnoides,the paper used the Hippophae rhamnoides juice residue as raw material extracted seabuckthorn oil,purited the unsaturated fatty acids,used microwave-assisted alkali method preparation conjugated linoleic acid.In this paper,the ultrasound-assisted extraction and the traditional extraction of Soxhlet extraction method were used for the Seabuckthorn oil extraction.In the ultrasound-assisted extraction,the ultrasound power,the time and the ratio of material and matliquid were considered.In the Soxhlet extraction,the time and the ratio of material and matliquid were considered.The optimization technology of ultrasound-assisted extraction was:360W ultrasound power,25min and the ratio of material and matliquid was 1:10,the yield of sea-buckthorn oil was 8.10 percent.The optimization technology conditions of ether Soxhlet extraction was:the ratio of material and matliquid was 1:20(g/mL),2h extraction.The optimization technology conditions of petroleum ether Soxhlet extraction was:the ratio of material and matliquid was 1:20(g/mL),2h extraction.The optimization technology conditions of N-hexane Soxhlet extraction was:the ratio of material and matliquid was 1:20(g/mL),3h extraction.The yield of seabuck-thorn oil on the optimization technology conditions were 14.5 percent,14.2 percent,13.2percent,seperately.The GC-MS result of Hippophae rhamnoides oils extracted by different methods showed:the major fatty acids composition were hexadecanolenin(palmitic acid),9,12-linoleic acid,9-palmitic acid,9,12,15-linolenic acid,oleic acid.The seabuckthorn fruit residue oils fatty acids extracted by different methods were not the same in the percentage of each component.The polarity of solvent was the main factor of impacted the oil yield extracted from the seabuckthorn.Ether Soxhlet extraction had best effect of unsaturated fatty acids,the proportion of unsaturated fatty acids reached 66.118 percent,which polyunsaturated fatty acids to 38.726 percent.The urea inclusion methord was used for purification of seabuckthorn polyunsaturated fatty acids.Polyunsaturated fatty acid included linoleic acid and linolenic acid in the original mixture the content from 32.33 percent to 78.14 percent increase,the best technical condition for inclusion was:fatty acids to urea ratio of 1:2,-6℃,16h inclusion.After purity the physical and chemical characteristics of the fatty acids was saponification value(mgKOH/g)201.12, iodine value(mg/g) 171.57,peroxide value(mmol/Kg)(POV) 1.83.Preparation conjugated linoleic acid through microwave-assisted Alkaline method. Inspected the microwave temperature,microwave time,the amount of catalyst KOH to fatty acid and the radio of fatty acid and solvent quality four factors impact of the linoleic acid into conjugated inoleic acid rate respectively.Used MINITAB program code design of the response surface experiment,MATLAB program quadratic polynomial regression equation fitting analysis,obtained the optimum conditions for conjugated linoleic acid by microwave-assisted Alkaline method was:microwave temperature 120.5℃,and the reaction time 10.8 min, catalyst KOH was 0.42 of the fatty acid,solvent 5 times of fatty acids in the amount,the conversion rate of linoleic acid into conjugated linoleic acid was 98.17%.
