Algae bloom is algae fulminic increasing on the condition of definite temperature and illumination,which making the color of water changed and forming colorful algae floater on the surface.The incidence of harmful algal blooms has recently increased in frequency as the water eutrophicated.The formation of harmful algal blooms has affected the health development of aquatic ecosystem.It is also harm for agriculture,water supply and travel business.Uptodate the main way to deal with the algae bloom are physical methods,chemical methods,,bio-methods and combinative methods of the above.The thesis has investigated the algacidal effect of antibacterial-deodorize textile,prepared antibacterial-algaecidal activate carbone and tested its algaecidal effect.to combine with the work of laboratory,nsp4 and nsp5 gene of rotavirus LLR were amplified,sequenced and their phylogenetic has been analyzed.1.Algaecidal effect of Antibacterial-deodorize textile Antibacterial and deodorize textile,as a textile which chelated cuprous and Basic Green,is effective to sterilize some normal prokaryotic or eukaryotic microorganisms.Algae-blooms causing algea are also prokaryotic or eukaryotic microorganisms.We investigated antibacterial and deodorize textile for algaecidal effect in the laboratory and experimental pool.It has showed that the concentration of chlorophyl a(Chla) of algal culture of Microcystis aeruginosa DS and Microcystis aeruginosa 7820,Plectonema calothrichoides Gom and Chlamydomonas reinhardtii had apparently decreased respectively after 3 days when the antibacterial and deodorize textile(0.4cm2·mL-1) was rinsed in the algal suspension cultured in the lab under 28℃and light intensity of 3000 lx.After 3 weeks of treatment, the concentration of chlorophyl a has decreased by 94.8%,92.7%,93.2%and 98.9 %,respectively for these algae.The Chla concentration of wild Microcystis aeruginosa algae in a pool(1.8m×2.8m×1.5m) of man-made algal bloom has decreased by 86.8% after 8 days treatment when the textile(1.25m2·m-3) was rinsed in the pool.Acute fish toxicity test(5 days) of the antibacterial and deodorize textile showed no significant toxicity.2.preparation and Algaecidal effect of Antibacterial-Algaecidal activate carbone Cu2+ has algaecidal effect,the antibacterial and algaecidal activate carbone were synthesized by coating polyacrylonitrile-phenylethylene and Cu2+ on activate carbone sueface,and tested the algaecidal effect of antibacterial and algaecidal activate carbone in the laboratory.It has showed that the concentration of chlorophyl a(Chla) of algal culture of Microcystis aeruginosa DS and 7820,Anabaena 7120,Plectonema calothrichoides Gom,Chlamydomonas reinhardtii and Scenedesmus obliquus K(u|¨)tz had apparently decreased respectively after 2 days when the antibacterial activate carbone (5g·L-1,10 meshes) was rinsed in the algal suspension cultured in the lab under 28℃and light intensity of 3000 lx.After 10 days of treatment,the concentration of chlorophyl a has decreased by 93.7%,96.0%,86.7%,94.3%,92.2%and 88.0%,respectively for these algae.Acute fish toxicity test(5 days) of the antibacterial and algaecidal activate carbone showed no obvious toxicity.3.Cloning and Phylogenetic analysis of nsp4 and nsp5 gene from Rotavirus LLR Rotavirus,one genera belongs to the family Reovoridae,known as the main cause of severe,dehydrating diarrhea in infants and young children.Since there have been no specially good effect remedy,the development and universal use of a safe and effective vaccine is considered as the first strategy for prevention.In this study,the nsp4 and nsp5 gene of rotavirus LLR are amplified by reverse transcription polymerase chain reaction and cloned into the plasmid pUCm-T,and was sequenced and analyzed,the nucleotide sequence of nsp4 and nsp5 gene has been shown highly identities(99%) with other lamp rotavirus.
