Lycopene is an important natural carotenoids,it has the function of various health cares,such as resisting cancer,increasing the immunity and deferring decrepitude etc.As a potential functional natural colorant,more and more people pay attention to it.The present work focus on pretreatment of raw material;the conditions of extraction , separation and purification; antioxidant activity;the development of microencapsulation and soft capsule; the stability of the product of lycopene. The results are as follows:We founded that freeze-drying was more effective than the other methods ,such as: ethanol dehydration, saponification, vacuum drying, ultrasound.When raw material's water content was less than 10% , the effect was 10 times than other methods.This article mainly dealed with the affecting factors,such us solvent,extraction temperature,time,pH,solvent and solvent ratio on extraction. And used orthogonality experiment to have ascertained optimum technological conditions.The results showed that extract of lycopene with 1:3ratio of raw material to the mixed solvent (acetone: hexane=3:1) for 4h in under T=35℃,extracted two times could obtain the best result.The study showed that the two methods have a very good separation of lycopene by ordinary chromatography and low pressure chromatography.The purity of lycopene reach to 95%,extraction rate reach to 0.30mg/g, which reach to a leading domestic level.We confirmed the structure of lycopene was all trans lycopene.the structure as follows:Produced two products of lycopene: microcapsules and soft capsules, and creat two production lines. This article also studied on the stability of the products,such as temperature, sunlight. The results showed that the products have very good stability to temperature and sunlight, when it was storaged in the -4℃for a month,the residual rate remained at about 90%, reach to the requirements of "the evaluation principle of health foods" .The antioxidant activity of lycopene was evaluated by hydroxyl radical and DPPH method. The results showed that when the concentration of lycopene on 40μg/mL , the hydroxyl radical clearance rate reach to 100%;the concentration on 100μg/mL, DPPH clearance rate reach to 64.5 percent.
