Surface Enhanced Raman Scattering spectroscopy(SERS) shows that high sensitivity for deterring the molecules adsorbed on the surface of the metals of surface phenomena, and can give the rich information of the molecular structure. With the in-depth understanding of the principles of SERS, the improvement of the stability of the laser and the sensitivity of the detectors, as well as the advances technological of the weak signal extraction and processing,the single-molecule detection sensitivity of the SERS spectrum of technology applications will be developed by leaps and bounds.The same substance can create different Raman enhanced effect on the different SERS active substrate. I use the Rhodamine B as detective molecule in this paper, the SERS spectrum from a higher concentration to the single-molecule level in silver solin.Use of surface-enhanced Raman scattering techniques study on Rhodamine B SERS spectrum,and identified the Raman peaks.Fluorescence and Raman spectroscopy for the same linear Gauss, fluorescence spectroscopy is broader than Raman. However, in the detection of Raman spectroscopy, fluorescence interference is inevitable. At single-molecule surface-enhanced Raman spectroscopy, the fluorescent interference is very strong, which interfere with the analysis of Raman spectra had an impact on, or even impossible to detect substances measured analysis. so that information extracted by the corresponding effective barrier, so filtered fluorescence is extremely important.In this paper,on the basis of Fourier transform principle,we imitate the test and use of digital filtering methods to filtered fluorescent interference, extract the Raman signal, and implementation of the single-molecule detection level.
