The production of steroid drugs and hormones is one of the best examples of the successful application of microbial technology in large scale industrial processes.The antitumor drug-exemestane is a useful aromatase inhibitor and is traditionally synthesized by chemical method. A lot of chemical poisonous agents are required and many by-products are produced.It is very difficult and ineffective to extract exemestane from these by-products.firstly,we studied the whole chemically synthesized processes of exemestane.Althougth a methylene group being introduced and modified in the androstenedione is easy and simple,it is very difficult for us to transform 6-methylenandrosta- 1,4-diene-3,17-dione to exemestane.The by-products are complicated and difficult to remove.Secondly,β-cyclodextrin(β-CD) and 6-methylenandrosta -1,4-diene-3,17-dione were used to form a complex.Theβ-cyclodextrin could notablely improve the water-solubility of 6-methylenandrosta -1,4-diene-3,17-dione and the complex has been tested by using some methods such as IR\UV\TG\DSC.The substrate can be successfully transformated to exemestane by Arthrobacter simplex.The work clearly showed thatβ-cyclodextrin complexation technique could enhance the exemestane production.In a lower substrate concentration,the exemestane production surpass 90%.We also set up the method of TLC and HPLC to quantitate the exemestane and related synthetic intermediates.
