The gust structure could affect the inclusion complexation and bio-transformation in cyclodextrin(CD)medium,but the rule is not clear.In this study,the biotransformation of 3-ketone steroidal ?~1-dehydrogenation reaction by Arthrobacter simplex TCCC11037 was selected as a model system,and a series of 3-ketone steroid compounds were selected for evaluating the effects of steroids structure on inclusion and the steroidal?~1-dehydrogenation process medium.The relationship between the steroid structure on inclusion and ?~1-dehydrogenation was established.This research will provide the theoretical basis and methodological guidance for the selection of substrate and the design of the catalytic reaction route in the supramolecular media.The apparent solubility and complexation stability of 3-ketone steroids in HP-P-CD medium were investigated by phase solubility method.The results showed that the apparent solubility of the 3-ketone steroids was linear with the increase of HP-P-CD concentration.When the structure difference was present in C-11,the solubilization multiple and the rule of complexation stability constants were C-11-hydroxyl<C-11-ketone<C-11-methylene,and they were consistent with the rule of the C-11 polarity of the compound.When the structure difference of 3-ketone steroids was present in C-17,the solubilization multiple and the rule of stability constants of complexation were consistent with the polar and size rule of the C-17 position.The ?~1-dehydrogenation of 3-ketone steroids in HP-?-CD medium was studied,the results showed that when the structure of 3-ketone steroids was difference in the C-11,the increase of the initial conversion rate was C-11-hydroxyl<C-11-ketone<C-11-methylene.When the structure of 3-ketone steroids was difference in the C-17,the increase of the initial conversion rate was consistent with the length rule of the C-17 group,and the longer of the side chain the more obvious of the enhancement effect.The relationship between complexation stability constants and steroids structures,as well as the relationship between initial conversion ratio and steroids structures were analyzed,the results showed that there was a negative correlation between the complexation stability constants and molecular size,and there was a positive correlation between the complexation stability constants and side chain length.At the same time,the side chain length has positive correlation with the increase of the initial conversion ratio,and the molecular size and molecular polarity have negative correlation with the increase of the initial conversion rate.
