There are two methods for detecting Deoxynivalenol (DON) and Ochratoxin A (OTA) in barely have been developed. An analytical study based on immunoaffinity column clear up, and HPLC for detection of DON in malting barley. Recoveries of DON from malting barley spiked at levels from 0.10μg/g to 2.00μg/g ranged from 92.04%-97.29%, with relative standard deviation between 0.92% and 2.67%. The limit of detection was 0.01μg/g. Of the total samples contained DON at levels ranging from 0.0123 to 0.0926μg/g.An analytical study based on immunoaffinity column clear up, and HPLC for purification and quantity detection of OTA in malting barley. Recoveries of OTA from malting barley spiked at levels from 0.10μg/kg to 10.00μg/kg ranged from 93.15%-97.07%, with relative standard deviation between 0.54% and 2.19%. The limit of detection was 0.05μg/kg.On the basis of two successful methods, the objective of this study was to investigate the variety of DON and OTA contamination during the process of brewing and the influence of DON and OTA on physical and chemical indicators of beer. DON and OTA were lost in the steep, lost 88.6% and 87.7%. However, DON and OTA were increased during germination and mashing. No change in DON and OTA levels was observed during kilning, boiling and brewing fermentation process. In addition, DON and OTA reduced barley germination and it developed low enzyme production and malt extract. The presence of DON and OTA reduced the kolbach index of protein dissolution,α-amino nitrogen and soluble nitrogen content, increased viscosity and diastatic power.Ozone detoxification method is a better method comparative results with many detoxification methods. malting barleys spiked DON and OTA are dealed with ozone produced by ozone generator, the concentration of ozone is 30mg/L, the treatment time is 60 min, the samples treated with ozone detected by HPLC. The results show that the effect of ozone detoxification method is good, it is not exist the problem of chemical residues, and will not affect the germination rate of barley.
