Tyrosinase (EC1.14.18.1), also known as polyphenol oxidase, is the key enzyme in mammal's melanin synthesis. Tyrosinase is a copper protein, which plays an important role in catalytic way. Lactoferrin is the protein witch has the properties of anti-bacterial.The purpose of this experiment is grafting LF onto wool, so that the wool has the properties of anti-bacterial.This paper investigated the best catalyzed oxidation condition of Tyrosinase from potatoes with simple and easy method with catechol(catecha phenol) as substrate. Then grafted wool which was catalyzed by tyrosinase with analog compound(sodium sulfanilate),then analysed with UV and FTIR-ATR. At last grafted wool which was catalyzed by tyrosinase with Lactoferrin, then analysed with UV,FTIR-ATR,SEM and Anti-bacterial. The experimental results show that: self-made potato tyrosinase has a certain vitality, the best conditions for the catalytic oxidation is temperature 30℃, pH 6.8, the stored days≤3 days. The best grafting efficiency is 45.33% when 200U/g and 30r(oscillation rate) by grafted sodium sulfanilate, then proved that the reaction was happened by UV and FTIR-ATR. One bath way can graft Lactoferrin by FTIR-ATR and SEM, the grafting efficiency is 36.8% at catalytic wool and 12.5% at non-catalytic wool by UV spectrum . The antimicrobial rate is 56.32% at catalytic wool and 12.63% at non-catalytic wool by antibacterial test. The best condition is pH9 and temperature 60℃by two bath way. And the antimicrobial rate is 43.68% at catalytic wool and 17.74% at non-catalytic wool by antibacterial test.
