In this paper,extraction process,initial decontaminate process and refined process of flavonoid of hawthorn leaves are researched.As a result,a rational process flow is obtained which lays the foundation for the industrialization of flavonoid of hawthorn leaves.Soxhlet extraction method is used to extract the flavonoid of hawthorn leaves.Based on the signal facts experiment,method of response surface experiment design is applied to optimize the extraction process.Net result:extractiong with 25g hawthorn leaves,the analog value of flavonoid extractionis 0.6718 g,medium value of experiment is 0.6710 g; the rate of flavonoid and the content of flavonoid respectely are 2.68%and 7.21%;the productive rate of crude extraction is 37.3%.With the initial decontaminate of extractive, the purity quotient of flavonoid is 24.0%,recovery is 66.8%.Macroporous absorption resins FL-1,FL-2,FL-3 and AB-8 and polymide are applied to refine the primary purification production,comparing the purification function of polymide,AB-8,FL-1,FL-2 and FL-3 with the absorption and elution experiment of static state and dynamic state,the result is that FL-3 and AB-8 have the highest recovery rate of elution and the best effect.Effective facts which influence the dynamic absorption and elution of FL-3 and AB-8 are experimented,and the optimal operational conditions are obtained.When sample size, velocity of leacheate,alcohol concentration,pH value of leacheate and velocity of elution respectively is 20.0 mg/g,1.0 mL/min,50%,6 and 1.0 mL/min,the purification effect of FL-3 is that purity quotient of flavonoid is 86.6%and product weight is 0.143g,recovery rate is 85.0%;When sample size,velocity of water,alcohol concentration,pH value of leacheate and velocity of elution respectively is 20.0 mg/g,1.0 mL/min,70%,6,1.0 mL/min,the purification effect of AB-8 is that purity quotient of flavonoid is 85.0%and product weight is 0.158g,recovery rate is 80.0%.In the paper,the purification effect of new ATPE of water-alcohol-inorganic salt and traditional ATPE of water-PEG-inorganic salt are researching too.The result is ammonium sulphate has the best parvafacies effect,so water-alcohol-ammonium sulphate and water-PEG-ammonium sulphate just are studied.Water-alcohol-ammonium sulphate systemy[alcohol:water:ammonium sulphate: primary purification production(mL:mL:g:g)=4:5:5:0.15]can enhance the purity rate of flavonoid to 50.9%,the recovery rate to 81.6%;water-alcohol-ammonium sulphate system 2[alcohol:water:ammonium sulphate:primary purification production(mL:mL:g:g)= 1:1:1:0.03]can enhance the purity rate of flavonoid to 47.6%,the recovery rate to 89.2%; water-PEG-ammonium sulphate system[PEG:water:ammonium sulphate:primary purification production(mL:mL:g:g)=6:5:4:0.15]can enhance the purity rate of flavonoid to 54.3%,the recovery rate to 93.0%.As we can see from above,water-PEG-ammonium sulphate system has better effect than alcohol:water:ammonium sulphate system,but the post treatment of PEG system is more verbose,and the price of PEG is higher than alcohol.
