Solanesol,a polyisoprenoid alcohol,which is an important pharmaceutical intermediate mainly being applied synthesis of coenzyme Q10 and vitamin K2 and the drug for cardiovascular disease and anti-tumor drug and anti-ulcer drug etc,has high application value.But,it is important in practical application of synthesis of coenzyme Q10 that the content of solanesol reaches above 90%.China,which there are lots of tobacco planting area,has already become the second major tobacco planting country,the most application and commercial value is synthesis of coenzyme Q10 with solanesol.This dissertation mainly concerned tobacco extract and synthesis and separation and purity of solanesol to gain high purity solanesol,to determine the optimum extraction process and operation parameters of solanesol and design the scheme and experiment of synthesis of coenzyme Q10.Organic solvent and supercritical CO2 were adopted to extract solanesol of tobacco.In according to the orthorhombic experiment and variance analysis,the optimal technological conditions were as follows for processing organic solvent extraction at a laboratory scale:extraction solvent was petroleum ether,extraction temperature was 60℃,extraction time was 6h;liquid -solid ratio was 9(mL/g),the number of extraction stage was 2,the extraction rate of PE was 15.32%,the content of the solanesol was 7.31%.In according to the orthorhombic experiment and variance analysis,the optimal technological conditions were as follows for processing supercritical CO2 extraction at a laboratory scale:extracting pressure was 20.0MPa, extracting temperature was 60℃,extracting time was 2h,CO2 flow rate 55kg/h, entrainer was 95%ethanol,the amount of entrainer was 1:1.5(g/mL),addition methods of entrainer was direct addition,the extraction rate of SE was 7.62%,the content of the solanesol was 9.38%.The results showed that Organic solvent extraction was effect.Saponification and solvent separation and column chromatography were adopted to separate high purity solanesol.The content of solanesol was 16.85%at the optimal Saponification reaction conditions.Water:95%ethanol=6:1(v/v) was adopted to wash soap stock,and to extract solanesol with petroleum ether,the content of solanesol was 36.30%.this method could omit dewaxing procedures and save cost. Methanol was adapted to separate extraction product of petroleum ether,the content of solanesol was 76.08%,the method would be effect and simple,and solvent recovery would be favorable too.By separation of column chromatography,the total yield of product was 68.20%.the content of solanesol was 94.85%in the products.Preliminary study on synthesis of coenzyme Q10 and synthesis of coenzyme Q10 of establishment with side chain extension method and side chain direct introduced method.Preliminary test of synthesis of decaprenol.
