Oligosaccharide has a good health role, the effectiveness of the industry that a new generation of food, a new type of functional sugar substitute for sugar source,and should have the prospect of a new product. Through the chromatographic separation method, a large number of oligosaccharide have been separated from the glucose production. However, the concentration of the oligosaccharide dilute solution is very low, only 5%, can not directly be used. The paper use sephadex gel in the concentration of oligosaccharides,because the gel extraction is a new separation technology with low energy consumption and not damage heat-sensitive materials, simple equipment and operation.Gel is easily renewable and can be reused, and it is conducive to energy conservation and reducing the cost.The gel extraction process in this paper adopts sephadex gel to improve the concentration of oligosaccharides. This paper is focused on the research of the synthetic methods and post-processing methods and reagents, and using gel extraction technology to concentrate the mother liquor ofoligosaccharides. The results show that the reaction temperature has a significant impact on the synthesis reaction, but the effect of concentration does not meet the expectations.Periodate was used as the oxidizing agent in the modification of sephadex gel. The effects of reaction temperature, reaction time and amount of periodate have been investigated. Response factors that affect the DS in size is amount of NaIO4> reaction time> reaction temperature. The DS can be over 0.84 under the optimum conditions that amount of NaIO4 is 0.05mol (the same molar of sephadex gel) and reaction temperature is 60℃, reaction time is 3h. Compared to the unmodified gel, the water absorption property of Sephadex gel can be greatly improved after the modification,and the absorbed water quantity of products can be Increased by 4~5 fold.Secondly, the paper use the modified sephadex gel to concentrate the mother liquor ofoligosaccharides. High efficiency liquid chromatography was used as the qualitative and quantitative analysis of the oligosaccharides dilute solution before and after the concentration. The effects of DS on the oligosaccharide concentration have been investigated. when the DS is 0.5 ~ 0.7, the increase in the concentration of functional oligosaccharide can be maintained at a relatively high level, up to 40%.Finally, the regenerated of modified sephadex has been studied, the regenerate ability rank is:K+< Mg2+< Ca2+. After compared with several regenerate methods, CaCl2 can be sure to be the ideal regenerant. The degree of gelation substituent will influence the regenerrate result of CaCl2, the rate of desorption is increasing with the raising of the substituent degree.But the rate of desorption approach to stability when the DS>0.6, and is up to 85% company with generally good regenerate result.What's more, with the increasing of CaCl2 concentration, the sephadex rate of desorption is raising rapidly but then slowed down gradually. The gel desorption rate achieve a higher level when the 0.1% in the concentration. With the increasing of the volume of regenerant, the desorption rate approximate to linear increase when the CaCl2 regenerant concentration is 0.1%, and the linearity coefficient is 0.9976.
