Lysozyme is a specialized enzyme for disruption of microbial cells which widely exists in the animals, plants and microorganisms with many advantages, such as perfect antibacterial performance, safe and non-toxic. As an ecological antimicrobial agent, lysozyme has been widely applied in the food, pharmaceutical, biotechnology and other industries and already been brought to the forefront in the textile industry. For instance, textiles can be provided with antibacterial performance when lysozyme is immobilized on them.At present, adsorption technique or crosslinker(glutaric dialdehyde) is researched in lysozyme immobilization mainly both at home and abroad. In this paper, Microbial Transglutaminase (MTG), a new biocatalysts, is employed to immobilize lysozyme on the wool fabric. It is not only a brand-new antibacterial technique, but also a new research idea in the wool modification.Based on the feasibility that MTG is able to catalyze crosslinking of lysozyme, this paper optimizes the immobilization conditions and researches the characterization of immobilized lysozyme. Finally, the antibacterial effect of the wool fabric immobilizing lysozyme was studied.The result studied by SDS-PAGE demonstrated that MTG is able to catalyze crosslinking of lysozyme for sure. Lysozyme can be immobilized on the wool pretreated by chemical agents, particularly potassium permanganate. The immobilizing conditions are as follows: 2g/L lysozyme, pH value 7.0, immobilizing for 8h with the MTG 20U/g.The result of the characterization of lysozyme indicates that the immobilized lysozyme has better pH and temperature stability. MTG-catalyzed lysozyme immobilized on the wool has better washing and operational stability than that immobilized by adsorption. The immobilized lysozyme catalyzed by MTG retains more than 80% of its original activity when it is stored in the dry condition after 30 days.The antibacterial finishing effect of the wool fabric immobilizing lysozyme shows that the inhibition percentage to staphylococcus aureus is 73.23%, however, there is no inhibition to Escherichia coli. The antibacterial effect after several washings of the immobilized lysozyme catalyzed by MTG is better than adsorption immobilization.
