Extraction, Purification And Biological Activities Evaluation Of Polysaccharides From Radix Actinidiae Argutae

The Radix Actinidiae Argutae is a kind of Chinese herbal medicines in common use, with a significant effect on tumour. Recently, various natural components have been extracted from Radix Actinidiae Argutae. Additionally, increased attentions focused on the development of the potential effects and market prospects of Radix Actinidiae Argutae.The polysaccharides of Radix Actinidiae Argutae (APS) are one of the mainly effective components ectracted from Radix Actinidiae Argutae. However, there is few report focused on the isolation,purifiation or biological activity analysis of APS.In this study, polysaccharides were extracted with boiling water and precipitated by alcohol. Proteins were removed by Sevag method and enzymolysis method. The optimal extracting conditions were concluded from a orthogonal experiment as follow: material and liquid ratio of 1:20, 100°C for 4 hours. The pre-experiment indicated that the alcohol addition is the determintive factor of polysaccharide precipitation. From the results, we found 3 times volume of alcohol is the best extraction addition. Then the products were further purified with column chromatography on DEAE-Sepharose Fast Flow and Sephacryl S-400, 3 fractions: APS I,APS II a and APS II b were obtained. The physicochemical characters of APS II a and APS II b were analyzed.Further, we examined the immunomodulation function and inhibitory effect of APS on tumor cell. The results indicated that the APS II b significantly inhibted HepG2 and SPC-A-1 proliferation with various concentrations. Additionally, the inhibtion ratio raised when the concentration increased. However, APS I and APS II a do not have obvious effects. It was concluded that APS II b was a polysaccharide which had the immunomodulation and anti-tumor function.In this study, the polysaccharides of Radix Actinidiae Argutae were extracted and purified, and the immunomodulation function and inhibitory effect on tumor cell of them were studied. It can provide a basis for further study of polysaccharides of Radix Actinidiae Argutae.
