Vitamin B2 is the composition of flavin mononucleotide and flavin adenine dinucleotide which are the coenzyme of flavin enzymes, In the biological oxidation process, it can play the role of transferring Hydrogen, and participating in metabolism of sugar, protein and fat in the body. It is long-term lack of vitamin B2 that the glossitis, stomatitis, mouth rot, eye fatigue, cataracts and other diseases will appear. Vitamin B2 can not be synthesized in the body, but need to be the exogenous food supplied. The grain is one of important sources of vitamin B2, therefore, to establish a simple, accurate and rapid determination method of vitamin B2 in grain has important practical significance.Currently, many methods with using fluorescence to determine vitamin B2 in vegetables, tablet were reported, however, the effect of the method with determining vitamin B2 in vegetables, tablet was used to extract vitamin B2 in grains was not satisfactory.The extraction of vitamin B2 has been improved and optimized in the text, and effect of the different extraction, extract method and the condition of extraction on VB2 in the millet was studied through single factor trial and orthogonal design trial. By adopting fluorescence luminosity law, the research measured content of vitamin B2 in millet onλex=444.0nm andλem=528.0nm. The result indicated that the quantity of VB2 extracted was higher when the experiment selected H3PO4 for extraction and used the water bath backflow. The effect of extraction was also better when extraction density was 0.1 mol/L, extraction time was 45 min and pH of solution was 5.0. And the text recovery reached from 89.20% to 100.32%, and the coefficient of variation was controlled by 1.21%. Hence, Precision and Nicety of this method was very fine. A new method for quick and exact determination of the VB2 in millet was offered.Under the effect of salt, water-soluble small molecule organic solvent can form two-phase system for extraction of target substances. Based on this a new method was put forward with isopropanol-salt-water aqueous two-phase extraction for extraction vitamin B2, Effect of different organisms, different organic amount, pH, and added a small amount of salts on the ability of separation phase of isopropanol alcohol was studied. The results showed that: under optimum conditions, one-time extraction rate of isopropanol-MgSO4 system to vitamin B2 was 97.6% to 98.4%, and the detection limit was 1.6×10-2mg/L. the relative error of the method used for the determination of vitamin B2 in rice was controlled by 1.22%. With the traditional liquid-liquid extraction and solid phase extraction compared, the method is simple, raw materials with cheap and abundant, interference with small, low toxicity, low power consumption and other significant features.
