Azadirachtin is tetracyclic triterpenoid, which is extracted from neem seeds and recognized by the world. It is used in agriculture, forestry and other role in biological insecticide. Azadirachtin have been separated from neem seeds, leaves, stems. They have a variety of biologically active substances and a wide range of medicinal value. Having are azadirachtin, nimbin, nimbidn, imbidol, nimbinate, ueceretin, salanmin so on. They have a role which is poor feeding, growth regulators, anti-shudder, anti-bacterial, anti-ulcer, analgesics, anti-arthritic, anti-fungal and so on. In this paper, extracting of azadirachtin has some reference value in development of biological agents.(1) Geting two identification methods of azadirachtin A, through accurate experimental testing,1, UV-Vis detection conditions and Standard curve of UV-vis detection.2, HPLC assay conditions and Standard curve of the HPLC.(2) Seven kinds of raw materials by HPLC (Bangladesh fruit, Myanmar fruit, domestic new neem fruit1, domestic new neem fruit2, domestic new neem fruit3, domestic new stone pillars3, domestic new neem pulp3) HPLC analysis showed that Myanmar fruit and Bangladesh fruit are different from its unique geographical environment and the atmosphere, Leading to d ifferent levels of azadirachtin. In comparison with domestic new neem fruit3, domestic new neem pulp3, The highest content of azadirachtin A is domestic new stone3, For these reasons, we choose the new domestic stone pillars3.(3) Because of the shortcomings of traditional extraction methods of azadirachtin A, which is long extraction time and low extraction efficiency, Other reason are solvent extraction waste and need for post-processing. In order to solve these problems aqueous, Proposed to solve these problems aqueous two-phase purification technology. The method is earring out in the physiological environment and not destroy the creatures living conditions, The entire operation is at normal temperature, in atmospheric pressure, under mild conditions equipment is simple, easy operation, the separation of fast and high recovery rate.The paper carried out a large number of single-factor experiments, in order to verify the feasibility of aqueous two-phase purification technology, The results showed that the concentration of inorganic salt was0.62g/mL, the extraction time was1.5h, and the material-solvent ratio was1:10(g:mL).By optimizing the range analysis of orthogonal experiment to use aqueous two-phase extraction and purification technology that Azadirachtin extraction rate determine the primary and secondary is Liquid ratio> extraction time> ammonium sulfate concentration. The extraction rate of azadirachtin was1.23%under the optimum condition that the concentration of inorganic salt was0.62g/mL, the extraction time was1.5h, and the material-solvent ratio was1:10(g:mL).(4) Under the same conditions, compared the orthogonal extraction, aqueous two-phase extraction purification with ethyl acetate. Ethyl acetate extraction efficiency is orthogonal, aqueous two-phase purification efficiency twice as much. Ethyl acetate is highly targeted and Can be extracted the material was90%one time.
