OBJECTIVEOptimize the extraction process of the activity parts of the medical materials of Salvia miltiorrhiza Bge. and Abutilon theophrasti Medic. Evaluate the quality primarily, in order to make a good basis on the development of Danqing soft capsule.METHODS(1) Optimize the extraction process of the seed oil of Abutilon theophrasti Medic. by the single factor and orthogonal design. (2) In order to primarily assess the quality specification of Abutilon theophrasti Medic., the following experiments have been accomplished:determine the physicochemical constant of the seed oil of Abutilon theophrasti Medic. according to the government standard; ascertain the best developing agent ratio and developer of TLC; primarily determin the chemical compositions by UV spectrum; determine the fatty acid composition of the seed oil of Abutilon theophrasti Medic. by GC-MS; investigate the stability of the seed oil of Abutilon theophrasti Medic.. (3) Optimize the extraction process of Salvia miltiorrhiza Bge. using the content of tanshinoneⅡA as a quota by the single factor and orthogonal design; primarily determine the chemical compositions by TLC; determine the main components of the liposolubility composition of Salvia miltiorrhiza Bge. by HPLC.RESULTS(1)For the factors'importance of extracting the seeds of Abutilon theophrasti Medic., the sequence is that the first important factor is soaking time, the second is solvent volume and the third is soaking times. According to the experimental results and practical production condition, the best extraction method is that take 10 times petroleum ether (60-90℃) to soak the seeds of Abutilon theophrasti Medic. twice in room temperature, and 36h each time. (2) The physicochemical properties of the seeds oil of Abutilon theophrasti Medic. include:the relative density 0.935; the acid value 1.54; the peroxide value 2.34 meq/kg; the saponification value 182.42; the iodine value 128.92. The best developing agent ratio of TLC is N-hexane:ethyl ether:ethanoic acid (80:20:1), and the best developer is 10% phosphomolybdic acid (ethyl hydroxide as the solution). The absorption maximum wavelength of UV spectrum is 210nm. After methyl ester of the seeds oil, eight types of fatty acids were determined by GC-MS. Including:linoleic acid 60.73%, palmitic acid 19.90%, oleic acid 11.48% and stearic acid 2.79%, myristoleic acid 0.39%, palmitoleic acid 0.42%, iso-oleic acid 0.55%. Of the total fatty acids the proportion of unsaturated fatty acids is 73.18%. After investigating the stability of the seed oil of Abutilon theophrasti Medic., we found that the oil was difficult to be oxidized in the condition of protecting from light or closing tightly. (3) The best extraction process is that extract the fat-soluble parts three times with 10 times dehydrated alcohol at 65℃, and 1.5h each time. And the best abstraction method with petroleum ether (60℃-90℃) is that extract three times with 10 times petroleum ether (60℃-90℃),and 25min each time. Detected the liposolubility active ingredients of Salvia miltiorrhiza Bge. by HPLC, and ascertained four main contents of the extracta sicca, as follows: tanshinone, tanshinoneⅡA, cryptotanshinone and dihydrotanshinoneⅠCONCLUSIONWe optimized the extraction process and primarily evaluated the quality of the activity parts of Salvia miltiorrhiza Bge. and Abutilon theophrasti Medic., which are the two medicinal substances in Danqing soft capsule, laying the foundation for the development of Danqing soft capsule.
