Meal is a kind of by-product by of plant seeds or core through squeezing or leaching method. Meal is rich in protein, starch, vitamin and mineral et al. If the meal is full used, it can help to decrease the pollution environment problem of greasyfood,meanwhile to improve the comprehensive development and utilization of meal.Thus it will be beneficial to increase the economic and social benefits, and greatly improve the development of meal resources.One strain, M1, which could degrade crude protein in meal, is successfully screened from the nature. Ml was preliminary as Bacillis sp. Using modern technology principle of microorganism fermentation, the sesame meal, soybean meal, rapeseed meal and teaseed meal were degraded by the Ml respectively, then the fertilizer efficiency of the four meals was compared through analysising of component in the meal after fermentation.The best suitable meal for Ml was choosed to ferment.Using this meal as substrate, Optimization the conditions of fermentation in shaked flask and the deep liquid fermentation were investigated,the results as following:The soybean's protein is the highest of 41.57% by analysing of the four kinds of meal.Afer fermentaion the degradation rate of protein is the highest about 73.80%.The content of amino acid is 23.84% in protein degradation product.In protein degradation process the loss of free ammonia nitrogen is 0.61%; and the organic acid conversion ratio is 0.18%.Thus soybean meal was choosed for fermentation substrate.The fermentation condition has been optimized by single factor and orthogonal experiments including the components of medium and the fermentation condition.The results showed that the best components of medium were the soluble starch of 1%, soybean meal of 8%,calcium chloride of 0.05%, potassium dihydrogen phosphate of 0.2% and the most suitable condions were the temperature of 35℃, pH 7, inoculation volume of 6%,rotation speed of 200rpm, fermention time 48 h. On these optimum conditions, enzyme activity can arrive at 507.61U/mL, biomass is about 12.7×108cfu /mL.Based on the study of liquid fermentation of shaked flask, the fermentation in 5L fermentor was experimented.The optimized conditions were temperature of 35 ℃, noculation volume 6%, rotation speed of 150rpm, ventilation volume is 250L/h.On these optimum conditions, enzyme activity can arrive at 687.25 U/ml, biomass is about 62×108 cru/mL.Compareing with the shake flask liquid fermentation,the enzyme and biomass have been greatly improved.
