Confocal Laser Scanning Microscopy (CLSM) is a kind of the laser optical technology with digital image processing technology, combining a high-resolution microscopy.Due to its advantages superior to the traditional microscope, CLSM developed rapidly in recent years. Compared with the traditional microscope, CLSM can scan the sample through the nondestructive detection in the natural environment, it also can give the 3D images of the surface,2D images of the section, CLSM has unique longitudinal resolution and high horizontal resolution, longitudinal and transverse resolution are 0.1μm and 0.001μm respectively.This thesis mainly forcused on observation and analysis of the basic method of eucalyptus wood chemical pre-processing preheated mechanical pulp (eucalyptus wood CTMP) in various RPM beating its surface physics, fiber surface and lignin and lignin and fluorescence intensity, cellulose and overall cellulose fibre inside the fluorescence intensity distribution and its actual wood grain, cellulose content relation curves, etc.Research results show that, by CLSM observation platform, with comments for Safranin O to carry on specific wood dyeing, Astra blue to specific staining through further experiments have found that can use two kinds of fibers are dyes and dye doesn't happen between chemical reaction, with the increasing number of beating turn, fiber surface fluorescence intensity of lignin and its total fluorescence intensity is weakened, and total fluorescence intensity of cellulose is increased, they both the trends and its actual testing content changes,Use the fiber analyzer FS-300 analyses different RPM of beating pulp, along with the increasing number of turn PFI beating the fiber average length, width, thickness and fiber surface fluorescence intensity (can characterize fiber surface content), the present reduce fiber surface roughness increases unceasingly, the degree of the fibers.
