Chlorogenic acid which is one of the phenylpropanoid syntheticed by the pentose phosphate pathway intermediates that produced by the process of aerobic respiration of plant, mainly reside in the Caprifoliaceae Lonicera, Compositae Artemisia and Eucommia. Numerous studies showed that chlorogenic acid had a wide range of pharmacological activity and biological activity, and were effective on the disease of digestive system, blood and reproductive systems. In modern study of biological activity, Chlorogenic acid had gradually penetrated into food, health, medicine and daily chemical industry and other fields.In this paper, chlorogenic acid extraction process was studied, the traditional method was improved in Qimen Huangshan honeysuckle. Single factor test on the major influencing factors such as extraction solvent type, concentration, pH, extraction time, extraction temperature, and solid to liquid ratio was studied. Then based on the single-factor test the orthogonal test was designed to optimize the extraction conditions which were concluded as follows:pH 4, the ratio of solid to liquid 1:20, the temperature was 60℃, 60% ethanol concentration, extraction time was 1.5 hours. Under this condition, the average extraction of chlorogenic acid was 3.67% and the purity of chlorogenic acid was 4.63% detected by HPLC.D101 macroporous resin column chromatography was used to purify semi-finished products of chlorogenic acid, by the single factor and orthogonal tests the main factors such as concentration of the elution, elution solution amount, pH, and elution speed was studied. The optimal elution condition was:20% ethanol eluent, eluent volume 5BV, eluent pH 8, elution flow rate is 4BV/h. Under this condition the purity of chlorogenic acid was 58.3%, yield was 76.9%.Further experiment was studied by Sephadex G-15 resin to purify Chlorogenic acid products of D101 macroporous resin. Through the experiment, the optimal elution conditions had been identified:50% acetone, eluent pH 4, elution rate 1ml/min. Obtained under these conditions the purity of chlorogenic acid was 82.52%, yield was 37.6%. After extraction with ethyl acetate, purity of products of chlorogenic acid reached to 86.16%.In the end, a process which had some practical significance for distilling and purifying Chlorogenic acid from Honeysuckle had established.
