Chlorogenic acid with the effect of antihypertensive is an important active ingredient of Eucommia, and gutta percha is an unique hard rubber of Eucommia, both of them are important substances contained in Eucommia. However, due to the cost of extraction and purification process, they have not been effective development. The market prospects of chlorogenic acid and gutta percha are broad, so the extraction and purification of chlorogenic acid and gutta percha had a very important economic significance.According to different dissolution properties, chlorogenic acid and gutta percha can be extracted by step approach. First water was used to extract chlorogenic acid, and the solvent was purified by NKA-9 resin; extraction residue was treated by alkali to decompositethe cell wall, and then extracted with petroleum ether, extraction solution was treated cool to get crude rubber, then got gutta percha by washing with acetone. So the chlorogenid acid and gutta percha were separated.Compared of different methods of UV spectrophotometry, paper layer-UV spectrophotometry, complex chromogenic reaction method, ferric chloride method and HPLC for determination of chlorogenic acid from eucommia ulmoides in precision, stability, reproducibility, and content determination results, and determined chlorogenic acid content in crude extract, The results meant these methods all had a high precision and reproducibility; the ferric chloride method had a lower stability, and a higher recovery rate; HPLC and paper layer-UV spectrophotometry were used to crude extract, sperm extract and pure solution of chlorogenic acid; paper layer-UV spectrophotometry had a lower impact of impurities, a higher accuracy, witch was an appreciation method for chlorogenic acid content quickly and accurately.On the basis of single-factor experiment of extraction temperature, extraction time, extraction times and solid-liquid ratio, designed a three factors and three levels of response surface experiment, the result of optimum conditions was,2 hours and 40 minutes of extraction time,81โ„ƒof extraction temperature,1:21 of solid-liquid ratio, the extraction rate of chlorogenic acid was 0.261%.Studied and optimized the purification process of chlorogenic acid with NKA-9, examined the effect of sample solution volume and flow rate on the adsorption, the choose of elution solution, eluent concentration and eluent volume on recovery and purity of chlorogenic acid, the result was,0.13mg/mL of the sample concentration,3 BV of sample solution, 2mL/min of the flow rate,4 BV of 10% ethanol of elution, collected fractions of high content of chlorogenic acid which was identified by mass spectrometry, got a purity of 35.8%,85% recovery rate of chlorogenic acid.Studied the extraction process of gutta percha from the residue, optimized the process of alkali treatment conditions, which was:10% NaOH solution treated in 3hours at 90โ„ƒ, with the solid-liquid ratio of 1:10; optimized the process of extraction with petroleum ether, which was: 2 hours extraction at 85โ„ƒ, with the solid-liquid ratio of 1:16; determined the cooling time which was 2 hours. With the optimized process conditions, got the extraction rate was 2.431%, with the purity of 85.6%.
