Tobacco aroma is integrated feelings of flavor and taste of physical sense, is the unique style and flavor of cigarette smoke. Aroma components of tobacco is closely related to smoke concentration, physical strength and aroma, and it's content is very trace though the chemical components in tobacco aroma is diversity and complex .Deficient of tobacco aroma is one factors of the tobacco quality and commercial value. With the growing concern of"smoke and health", developing lower tar cigarette has become the trend for survival and development of tobacco industry. Reducing tar of cigarette may inevitably lead to decline the amount of cigarette aroma, which can be supplemented, modified and improved by addition of the tobacco additive of flavor and fragrance in later stage.There have a number of preprocessing methods used for collecting the aroma components of tobacco: solvent extraction, water stream distillation, solid-phase micro extraction, headspace analysis, simultaneous distillation extraction and supercritical fluid extraction and so on. In this paper, the aroma components of tobacco were got by ultrasonic-assisted solvent extraction, simultaneous distillation extraction and dynamic headspace analysis (purge & trap), the aroma components were determined qualitatively and quantitatively by gas chromatograph/flame ionization detector/mass spectrometry (GC/FID/MS), quantitative result of different types of tobacco was statistically analyzed by included angle cosine and Euclidean distance combined, result was satisfactory. Main researched contents and productions were summarized as follow:1. Simultaneous distillation extraction (SDE),ultrasonic-assisted solvent extraction (ULSE), purge and trap(P&T), liquid-liquid extraction(LLE), micro liquid-liquid extraction(MLLE) and direct injection preprocessing method were compared for their effectiveness in the extraction of aroma components in tobacco and tobacco essence, influenced factors of the these methods to result of quantitative and qualitative of aroma components had been discussed. Experimental results showed that ultrasonic-assisted solvent extraction (ULSE) and simultaneous distillation extraction (SDE) were suitable for proper quantitative and qualitative analysis, while dynamic headspace (P&T) was suitable for rapid qualitative analysis, LLE was suitable for routine analysis and quality control of tobacco essence as the first preprocessing method.2. A feasible method which is similar to two-dimensional chromatograph(GC+GC) had been developed for tobacco aroma components by studying of MS vent terms and various polarity column, the result of precision and recovery of this method were satisfactory.3. Quantitative and qualitative results of tobacco aroma components of different region and grade were evaluated with similarity analysis by included angle cosine and Euclidean distance combined, it provides an effective means for the comprehensive evaluation of tobacco quality.
