With the problem of global energy shortage and environmental deterioration become more and more seriously, people are exploring the renewable energy to substitute the fossil fuel. Biodesel is such a kind of clean and renewable energy. The raw materials of biodiesel are vegetable oil and waste oil at present. However, the high material price of vegetable oil and waste oil led to the high production cost of biodiesel production. So develop cheep materials are very important for the biodiesel industrialization. Some studies find that many microbe have a great deal of oil in the body and can co-fermentate glucose and xylose simultaneously, so production of oil material by microbial fermentation using hemicellulosic hydrolysate product can provide a wide range and cheap raw materials for biodiesel.The UV and DES were applied as compound mutagenesis to study on protoplast mutation from Trichosporon cutaneum GIM2.68.The result indicated that when the UV lamp is 30W, the Irradiation distance is 30cm,the Irradiation time is 60s, the mutation strain (pi-60) can be selected.The oil and biomass of the strain pi-60 could be respectively reached up to 30.4% and 1.1354g/100ml.When the DES were applied to the strain pi-60, the mutation strain (pi-60-70) can be selected under the treatment concentration of DES is 1%, the treatment time is 70min. The oil and biomass of the strain pi-60-70 could be respectively reached up to 35.7% and 1.2573g/100ml, compared with the starter strain,the content of oil has improved 33.7%, The biomass has improved 66.7%.The acid, enzyme, acid and enzyme, alkali and enzyme, microorganism were used to treat corn stalk. The best process conditions were obtained by orthogonal test through measuring the content of reducing sugar in product. When Cellulomonas flavigena was used, the additive of Cellulomonas flavigena was 5%, the cultivated temperature was 31℃and the cultivated time was three days. The result shows that the content of reducing sugar in product was 5.720mg/ml. When Cellulase and Trichoderma koningii were used, the additive of Cellulase was 0.15%, the additive of Trichoderma koningii was 10%,the cultivated temperature was 30℃and the cultivated time was six days. The result shows that the content of reducing sugar in product was 4.326 mg/ml.The fermentation product of corn stalk were used as main material to culture pi-60-70, the best process conditions by orthogonal test through measuring the content of oil in fermentation product were that the reducing sugar concentration was 4.326 mg/ml, the additive of pi-60-70 was 11%,the cultivated temperature was 28℃and the cultivated time was seven days. The result shows that the content of oil can reach 3.321g per 100g fermentation product.
