Enantiomerically pure Cyanohydrin,enolase and propargyl alcohol are important chiral intermediates in organic synthesis. They therefore provide a wide space for transformation into a large number of chiral molecules, which are important in the synthesis of biologically active molecules. In this dissertation, a study on the stereoselective formation of these two kinds of compounds by kinetic resolution (KR) has been performed. The study was KR of Cyanohydrin,enolase and propargyl via enantioselective acylation.(1) Synthesis of 2,3-dihydroimidazo compounds as catalystsFrom cheap starting materials readily available to the higher yield synthesis of two 2,3 dihydro-imidazole-thiazole core acyl transfer catalyst.(2) Enzymatic kinetic resolution of cyanohydrinsBy using 2,3 - dihydro-imidazole catalyst, kinetic resolution of a series of racemic cyanohydrins has been achieved via stereoselective acylation. Research shows that benzene replacing aliphatic cyanogen alcohol, because benzene from the reactive center is farther, strengthened and active catalyst formed between cation -Ï€force, which greatly improve the reaction of the chiral selectivity, s value for 111, short carbon chain aliphatic cyanogen alcohol if aromatic ring replacing, benzene can promote the substrates and catalyst interactions. The reaction conditions were optimized. Substituent effect was also briefly discussed.
