The extracting technique was optimized for the extract from the stems and leaves of P.edulis f. flavicarpa (PEFE) which was proved to have anxiolytic activity. The extracting recovery of flavonoids, the important neuropharmacological active substance, was used as the index to assess the extracting technological conditions. Firstly, quantitative determination method of total flavonoids was established; next, the effects of the technological factors including the concentration of ethanol aqueous solution, extracting method, the ratio of solvent to stuff, and so on, on the extracting recovery of flavonoids were investigated by orthogonal design. Finally, the extraction processes for PEFE were determined as follows:the model of three-stage countercurrent extraction for three times was adopted, in each extraction, humidified raw material with 2 times of 50% ethanol aqueous solution with was refluxed with 10 times of 50% ethanol aqueous solution for 0.5h.For the PEFE prepared according to our extracting technique, quality standards were established. Referring to European Pharmacopoeia 5.3, qualitative method of PEFE was established by thin-layer chromatography after colored with Al3+; quantitative method of PEFE was established by determining the content of total flavonoids and isoorientin (the high-content compound in the stems and leaves of P. edulis f. flavicarpa with neuropharmacological activity). The content of isoorientin was determined by HPLC and the method was proved to be specific and accurate with wide linear range, the sample solution was stable in 10 hours. The contents of total flavonoids and isoorientin were determined in six batches of PEFE. Based on the measurement results, the content of total flavonoids in PEFE was set as no less than 2% and that of isoorientin no less than 0.5%. The loss of water and ignition residue of the 6 batches of PEFE were assayed and the standards for the two indexes were set as no more than 5% and no more than 10%, respectively.Because leaves of P. edulis f. flavicarpa presented anxiolytic activity while leaves of P. edulis f. edulis showed sedative activity on high dose in our previous work, we compared the flavonoid compositions, the important neuropharmaco-logical substance in plants of genus Passiflora, between the two forms of P. edulis by RP-HPLC. The chromatograms of the samples belonging to similar population of Passiflora edulis were identical, but those belonging to different population were distinct from each other. The series of peaks between 16 and 24 min in the chromatograms of Passiflora edulis'flavicarpa'did not appear in those of Passiflora edulis'edulis', either did the peaks between 54 and 90 min in chromatograms of Passiflora edulis 'edulis'not appear in those of Passiflora edulis 'flavicarpa'. The six major flavonoid compounds isolated from the leaves of Passiflora edulis flavicarpa', lucenin-2, vicenin-2, isoorientin, isovitexin, luteolin-6-C-chinovoside, and luteolin-6-C-fucoside, had not been detected in Passiflora edulis 'edulis'.The flavonoid composition and pharmacological effects of P. edulis f. flavicarpa were compared with those of P. incarnata reported by other researchers and the conclusion was made that P. edulis f. flavicarpa could be used as a resource of Passion Flower Dry Extract.
