Auricularia auricula is the Chinese traditional edible mushroom, which were reported to have many activitive compounds, including exopolysaccharides(EPS) and adenosine. From the perspective on that Chinese Herbs can inervente anabolism of edible-medicinal fungi produts, based on research in some typical regulation of fungal key metabolites by Chinese Herbs, extracts of Chinese Herbs added into medium.The yields of EPS and adenosine can be influenced, so different amounts of extraction of Chinese Herbs were adjusted in order to increase the concentration EPS and adenosine. Meanwhild, fermented Chinese Herbs by edible fungi can change pharmacology of Chinese Herbs, reduced side effects.In this paper, the content of EPS was chosen as indicator, four Auricularia auricula strains with high-producing EPS were screened, and the targer strain’s fermentation process was optimized. Under this conditions, the extracts of Acanthopanax senticousus, Glycyrrhiza uralensis and Schisandra chinensis were added to the medium broth to detect the dynamic changes of EPS and adenosine. Furthermore, Acanthopanax senticousus, Glycyrrhiza uralensis and Schisandra chinensis were fermented by the target strain and the contents of active compounds were determined in the fermented products. In conclusion,The four high market liquidity strains were selected, yield of EPS choosed as index to screen the high producing strain. The results showed that the Hei2011 strain was EPS high-producting strain with its yield was 4.081±0.018g/L. So the Hei2011 was chosen as target strain.Single factor tests and orthogonal experiment design were used to optimize the fermentation process of Hei2011 with the yields of EPS as marker. The best condition: the fungus age was 6d, the added volunm of suspension was 8%(V/V), tempreture was set at 28℃, the fermentation time was 6d, the speed of shaker was 140r/min. Under optimited condition, the EPS yield of Hei2011 was 4.550±0.023g/L.The effects of water extracts of Acanthopanax senticosus, Glycyrrhiza uralensis and Schisandra chinensis on the submerged culture of Hei2011 were studied. By adding the extracts of Acanthopanax senticousus was 4g/L, the maximum yields of EPS reached 6.022±0.018g/L and the maximum yields of adenosine arrive at 2.5044± 0.0129mg/g when 6g/L extracts of Schisandra chinensis was added..In order to study the change of the main active compounds in fermented Acanthopanax senticousus, Glycyrrhiza uralensis and Schisandra chinensis by Hei2011, the contents of syringoside and eleutheroside E in Schisandra chinensis, the contents of liquririn in Glycyrrhiza uralensis and the contents of lignan ingredients in Schisandra chinensis were detected, and the results showed that there were a significant reduction in sytingin and eleutheroside E contents in Acanthopanax senticousus, a little change of contents of liquiritin in Glycyrrhiza uralensis and greatly increase of schisandrin, schisantherin and γ-schisandrin contents in Schisandra chinensis.The studies showed that yields of Auricularia auricula EPS and adenosine were increased in different levels by adding extracts of Auricularia auricula and Schisandra chinensis to medium, but the results varied when added extracts of Glycyrrhiza uralensis. During Acanthopanax senticousus, Glycyrrhiza uralensis and Schisandra chinensis were fermented by Hei2011, some activ ingredients could be converted into another substances, while the contents of some acrive conpounds were increased..
