| In this paper,preparation of Chlorogenic Acid (CA) production process from the Eucommia leaf has been studied.The HPLC as the accurate method for analysis the CA which chromatographic conditions were decided.The analytical conditions of the Thin-Layer Chromatography-Ultraviolet spectrophotometer were studied which as the rapid analysis of chlorogenic acid method.The extraction conditions were optimized in warm water which added in cellulose for CA extracting,under the condition,the transfer rate remained stable at 92%.The separation process of the CA extraction has been discussed and the results showed that the extraction impurity and concentrated by membrane improved the adsorption capacity of resin significantly,as well as purification effect except resin pollution.The experiment noted that membrane process for adding enrichment could significantly increase the efficiency of resin.The effects of different membrane process for separation and purification has been studied.The concentration which from extraction precision-filter after impurity through micro-filtration membrane II obtained by the nanofiltration membraneⅡwas the optimum route.The process impurity removal rate 41.6%,The CA transfer rate of 83.2%.The appropriate microfiltration and nanofiltration operating conditions were determined over film studied membrane parameters and operating conditions,the micro-filtration membrane II under the conditions of the average flux was 253(L·h-1·m-2),The CA transfer rate was 95.6%.As well as the operating parameters of nanofiltration membraneⅡwere studied..The resin purification conditions of the CA concentrated solution from membrane were studied.After study the HPD400A resin was for further testing,the best of resin technology parameters and operating conditions were confirmed through factor analysis,The CA content which eluted by the volume fraction of 30% ethanol through vacuum distillation after drying between 56.9%~58.3%.This study established a process for preparation of CA in eucommia leaf through using membrane to impurity removal and concentration for extracted liquid before purified by macroporous resin.Through the process the yield and purity of product each was 87.9% and 57.5% also the emission of wastewater reduced by 90%.Filtrate was recycled, the pollution was controlled in the production, resulting in cleaner production. |
