Series researches of Eucommia ulmoides oliv were studied in this dissertation, HPLC analysis of chlorogenic acid and rutin’s were established. The technology conditions of chlorogenic acid and rutin from eucommia ulmoides oliv using ionic liquid and microwave-assisted extraction were optimized by response surface experiment, The microscopic structure changes of eucommia ulmoides oliv cells at before and after extraction were observed by using the electronic scanning microscope, and the mass transfer mechanism of ionic liquid and microwave-assisted extraction were analyzed. The purification technology of chlorogenic acid and rutin using macro porous resins was optimized by orthogonal experiment. Conclusions provided scientific foundation for utilization of eucommia ulmoides oliv.In the present work, the study results can summarized as following aspects:(1) HPLC analysis of chlorogenic acid and rutin were established. The baseline separation of chlorogenic acid and rutin was obtained under the chromatographic conditions. The linear equation of chlorogenic acid was Y=1306941.7367X-13775.0000, and correlation coefficient was0.9997. The linear equation of rutin was Y=2171691.9861X+42213.0000, and correlation coefficient was0.9995.(2) The technologies of the ionic liquid and microwave-assisted extraction of chlorogenic acid and rutin from eucommia ulmoides oliv were optimized by response surface experiment. Three kinds of ionic liquid,[Bmim]Cl,[Bmim]Br and [Bmim] PF4, were synthesized, and their structures were confirmed by IR.[Bmim]Cl was good solvent extraction of chlorogenic acid and rutin. The results showed that the appropriate technology conditions of [Bmim]Cl ionic liquid and microwave-assisted extraction of chlorogenic acid and rutin were optimized by response surface experiment as follows:Ionic liquid concentration2mol/L, the ratio of liquid to solid15:1(mL/g), extraction temperature50℃, extraction time10min, the yields of chlorogenic acid and rutin were1.58%and0.206%, respectively. Compared with microwave extraction, ionic liquid microwave-assisted extraction posessed shorter extraction time and higher extraction efficiency.The microstructure changes of eucommia ulmoides oliv cells at before and after extraction were observed by using of the electronic scanning microscope, and mass transfer mechanism of ionic liquid and microwave-assisted extraction was analyzed. The results indicated that different extraction ways had different effects on cell organization. Ionic liquid microwave-assisted extraction made cell wall rupture more easily and lowered the mass transfer resistance.(3) The purification technologies of chlorogenic acid and rutin by macro porous adsorption resin were investigated. S-8macro porous adsorption resins were used as adsorption carriers for the separation and purification of chlorogenic acid and rutin. The optimized conditions for the separation and purification of chlorogenic acid by orthogonal experiments was:50%ethanol to eluent300mL, agent velocity3.0mL/min, column liquid on pH value of1.0, and the purity of chlorogenic acid reached78.42%; The optimized conditions for the separation and purification of rutin by orthogonal experiments was:70%ethanol to eluent300mL, agent velocity2.0mL/min, column liquid on pH value of5.0, the purity of rutin reached56.41%. So ionic liquids microwave-assisted extraction can improve rate, and have a distinct advantage.HPLC of chlorogenic acid and rutin’s contents were established in this study. The technology conditions of chlorogenic acid and rutin by ionic liquid microwave assisted extraction were optimized, which provided a new way of sample preparation for chlorogenic acid and rutin. The conclusions provided basis and foundation for quality control and utilization of Eucommia ulmoides oliv.
