With the development of environmental science, there are many disadvantages in analyses method such as culture and it can't follow the development. Many studies in microbial ecology have focused on decribing the diversity of microbial communities and the relation abundances of the most commen member.In this study, denaturing gradient gel electrophoresis (DGGE) and fluorescence in site hybridization (FISH) with oligonucleotide probes were used to investigate microbial community composition in activitied sludge of metal membrane biology reactor.Metal membrane is becoming a new technology in High Load Domestic Wastewater treatment, it has a bright future. In the lab, the deep research work I focus on is the start and running of this system with nitration tank and both nitration tank and remove nitrogen tank. During the 114 days stable operation, the removal of CODcr was maintain about 85%, the removal of TN starts from 30% up until 90%, the removal of NH4+-N was leveled off at 90%, Water Quality:COD was about 10 mg/L, TN was down from20 mg/L to4 mg/L, NH4+-N was under 1 mg/L, NO3--N was down from 25 mg/L to 3 mg/L, NO2--N was reduced from 0.3 mg/L to 0.05mg/L, lower than the city wastewater comprehensive discharge standard.In the paper, I have done experimentation of metal membrane system to facilitate the process of experimentation, waste water simulating of sewage water was made by us to remove unnecessary disturbance. In the process of metal membrane system, DGGE picture revealed that there were some changes in bacterial community. PCR,DGGE,FISH molecular biological techniques were applied in the nitrification bacterium microbial communities, and discovered the relationship between the nitrification bacterium and Nitrogen Changing. The result of PCR-DGGE indicatec that:with the development of operation, the number of Microbial Community was adding. Aboundance was also changing.
