| The fluorescence microscope is used widely in a lot of fields of biology and medical science, now it has already become a kind of important and essential means for observation and test. With the development of biomedicine the performance of the traditional fluorescence microscope used extensively can't satisfy the demands. For example, the excitation spectrum is discontiguous, we can not get digitized fluorescence microscopic image information and spectrum information at the same time. So it is very meaningful to increase functions of the traditional fluorescence microscope aimed at its function defects so as to meet the needs of modern biomedical development and make it play a greater role.The principle of the continuous spectrum analytic system of micro- fluorescence image studied in this paper is rebuilding the existing fluorescence microscope. The structure of the fluorescence microscope is not destroyed. Through the new fluorescence microscope, we can get digitized fluorescence microscopic image information and spectrum information at the same time.This paper includes the following research contents mainly:1. Design of the grating monochromator controlled by the computer.2. Method for connecting the grating monochromator controlled by the computer and the fluorescence microscope.3. Method for connecting the grating monochromator controlled by the computer and the xenon lamp.4. Computer software design to control the monochromator to scan.5. The technical research of image preprocessing and the software design.6. The research of the methods of spectral analysis and the design of the continuous spectrum analytic software of micro- fluorescence image.7. Set up a set of experimental provision of the continuous spectrum analytic system of micro- fluorescence image.In this text, excitation spectrum scans by xenon lamp and monochromator, digitize fluorescence microscopic image by CCD, image acquisition card and computer, control the system and acquire process and analyse image by the system software designed by myself. This system not only remedied the function defects of the traditional fluorescence microscope, but also realized the computer control and the process and spectral analysis of the fluorescence image. The research of this system can offer us the theory and experiment experience of the upgrading of the traditional fluorescence microscope used widely in scientific research institution, such as hospital, police, material, etc. |
