Cryptosporidium and giardia are two common species of pathogenic protozoan, seriously endangering the water quality. It is very important to investigate the occurrence of cryptosporidium and giardia in water and wastewater since they threaten the safety of drinking water and bring healthy and ecological risk to city water system.In order to found effective method for detection, Problem group cooperated with Shanghai Pudong Veolia Water Co. Ltd. Method EPA1623 for monitoring and analysis cryptosporidium and giardia in Drinking Water. Problem group did a research about matrix spike/matrix spike duplicate, initial precision and recovery, process spike recovery. Concentration and separation, Immunomagnetic separation, immunofluorescence assay, three main steps in method 1623, were evaluated. Examination of the recovery of each step in this procedure indicates that Concentration and separation is the main recovery limited step.
