In this paper, we discuss the automatic improvement process of common microscope in detail.In order to realizing the auto-control of the microscope, the text starts in the fundamental structure of the microscope and carries through improvement bringing from the facts about the carrier, adjustment mechanism of the imaging lens and image acquisition device and so on .At the same time, we discuss about the problem on view field alignment and present the improvement way about the objective lens converter.Have recollected development of microscope first, the research introduces the relevance technology among the reforming and sums up progresses of auto-control microscope at home and abroad. The article has been discussed that it adopts step-by-step electric motor to come to auto-control through reforming the carrier, the design of the image acquisition device, from view systematic optical quality and demands to start off discussing to select and use CCD, adjustment mechanism of the imaging lens is designed by adding the step-by-step electric motor pretending to realize high foucusing accuracy and brings forward innovation design about the auto alignment device in micro field of view.In discussion that image clarity-evaluation-function, the article has adopt gray-level gradient operator to be the main object of study to make balance on gathering real time nature, calculating amounts, focusing on sensitiveness .At the same time,itadopts the Laplace square operator as the image clarity-evaluation-function, by collecting 20 different articulation human body blood photographs to carry out verification. The experimental results show that the algorithm is feasibility. A new search arithmetic on the focusing location is brought forward, though setting up capture image quantity in procedure, can change the size focusing on graduation, so that it can realize the accuracy of the adjustment mechanism of the imaging lens voluntarily right away.The article has been discussed the process program design of the system. Based on View for Windows (VFW), comments on image capturing program in detail and under the control of to step-by-step electric motor monolithic machine are discussed. In the last chapter, it carries out analysis on entire systematic accuracy, and draws the back-off scheme.
