Solanum nigrum L.has important medicinal value,edible value and commercial value. It has broad prospects for development and utilization. In this study, Daqing City, Heilongjiang Province Zhaozhou Solanum seed as the test material. They were experimentally manipulated to examine effects of basic medium, hormone type and concentration, explant type, Different environmental factors at the stage of adventitious buds induction, Adventitious buds proliferation and rooting culture as studied,and the effect of acclimatization time and transplantion medium on transplant was also studied. As a result, the best mediums of every stage were screened out, tissue culture system for S.nigrum L was established.Then explore plant growth regulators, basic medium, carbon sources and light application time which affected in vitro flowering were explorated. The main results as following:1.Establishment of in vitro regeneration system of SolanumSeeds were used to study in vitro propagation of Solanum, The best medium for adventitious bud induction was MS medium with addition of 3.0 mg/L 6-BA, 0.3mg/L NAA, for the leaves was a suitable explant type. for the Sucrose is the suitable carbon source, for propagation was MS medium with addition of 2.0mg/L 6-BA, for rooting was 1/2MS medium with addition of 0.5mg/L NAA, with the rooting rate of 99.67%. The best time of hardening plantlets was 3 d. When the plantlets were transferred onto the garden soil, and the survival rate was up to 93.33 % at proper temperature, and the Test tube seedling growth is very good, can grow.2.In vitro flowering of SolanumThe affecting factors of in vitro flowering were studied on the establishment of efficient regeneration system. Based on the results, the in vitro flowering rate of Solanum had little difference between MS and 1/2MS basal media. the in vitro flowering rate of Solanum had big difference between 6-BA and KT basal media. When the concentration of 6-BA was 1.0mg / L, the in vitro flowering was the best, and the flowering rate was up to 30.55%.Single use NAA can promote Solanum vitro flowering; The in vitro flowering was promoted under the light of 12h/d, Carbon sources also played an important role on in vitro flowering in Solanum, When the concentration of sucrose was30 g/L the the flowering rate was the best.
