High Frequency In Vitro Regeneration System And Vitro Flowering Of Potted Chrysanthemum With Small Inflorescences

The experiment took the petals of four varieties of Chrysanthemum as explant to establish high-frequency regeneration system based on petals of Chrysanthemum.The optimal hormone formulation of callus induction and adventitious bud differentiation was screened out by orthogonal experimental method,MS as the basic culture medium,adding auxin and cytokinin to system.Regulating regulation of vitrification by improving osmotic pressure,and reducing concentration of cytokinin and humidity of culture bottle as to vitrification phenomenon.The in vitro flowering was induced by flower buds of small potted chrysanthemum.Studying effect of different growth hormone on Flowering of plant in test tube through screening test of single factor.The results as follows:(1)Varieties of small potted chrysanthemum are ’Chrystal Pink’,’Rainbow Worth’,’Chrystal Bronze’ and ’Little Rock’,the formulas of callus differentiation media are MS+NAA1.2mg/L+6-BA 1.0mg/L+TDZ0.4mg/L,MS+NAA 1.2mg/L+6-BA 1.5mg/L+TDZ0.2mg/L,MS+NA A0.3mg/L+6-BA 1.0mg/L+TDZ0.4mg/L and MS+NAA1.2mg/L+6-BA 1.0mg/L+TDZ0.4mg/L respectively.The induction rates are 74.56%,77.21%,62.23%and 28.87%respectively.(2)Optimal induction medium of adventitious bud of ’Chrystal Pink’,’Rainbow Worth’and’Chrystal Bronze’ is MS+6-BA1.0mg/L+NAA0.1mg/L.Optimal induction medium of adventitious bud of’Little Rock’ is MS+6-BA1.0mg/L+NAA0.3mg/L+TDZlmg/L.(3)Optimal mediums of transgenerational proliferation of’Chrystal Pink’,’Rainbow Worth’,’Chrystal Bronze’ and ’Little Rock’ is MS+6-BA0.6mg/L+NAA0.05mg/L+TDZ0.1mg/L.Cytokinins of Optimal mediums of transgenerational proliferation of ’Chrystal Pink’ decreased by 50%and Vitrification rate of’Chrystal Pink’ decreased by 21.11%.Analyzing two factors of vitrification rate and increment rate comprehensively,It’s beneficial to improve added value of glass seedlings by increased by 50g/L about concentration of sucrose.(4)Optimal medium of take root of ’Chrystal Pink’,’Rainbow Worth’ and ’Chrystal Bronze’ is MS+NAA0.2,Optimal medium of take root of ’Little Rock’ is MS(5)Abloom rate is high and flowering phase is long when concentration of sucrose is 50 g/L in the medium.The abloom rates are 52.23%-65.56%,17.76%-28.89%,31.11%-37.78%and 39.89%-43.23%respectively by adding NAA,6-BA,TDZ and Kt of 0.1mg/L,O.lmg/L and 0.5mg/L.When add GA3 of 1.0mg/L to MS,the abloom rate reach to 39.89%-43.33%.
