The callus and hypocotyl were transformed by using Agrobacterium Tumefaciens and micropjectile bombardment, Study on every influencing factor. The stable and high performance system was established, the result was positive by Gus (B-Glurcuronidase) enzyme activity test and PCR (Polymerase Chain Reaction) test, which the foreign gene EFE (Ethylene Formation Enzyme) had been integrated and expressed in carnation was verification. This research result indicated: I .The basal medium suitable for carnation is MSZ+2.4-D 0.lmg/L+BA 0.2 mg/L MSZ+BA 0.5 mg/L+NAA 0.2 mg/L: l/2MS+IBAI.0 mg/L+GA3O.lmg/L+AC 0.8 mg/L0 2.In the method of co-cultivation , the best in fection concentration of Agrobacterium Tumefaciens is lox , and the best co-cultivation time is 3 days , and the best material is callus 3 .The best parameters of transgenesis by microprojectile bombardment is microprojectile bombandmenot velocity is I IOOpsi. Sample height is 6 centimeters 4.The receptors by the two different methods all possess the resistant of Km ,Which grew well on medium ,and can differentiation the plantlets, and reproducted the plant. 5.The foreign gene(EFE) was transferred into carnation varieties callus and hypocotyl successfully under the above mentioned moderate conditions.
