Three kinds of anticoagulant were used in the blood infected by Eperythrozoon ovis .The result indicated that Citrate affected it's multiplication most in vitro. And Eperythrozoon ovis increased by add glucose and NaHCO3 in citrate.Blood sample must be treated within 12h after collect from the body.Epidemiology investigated through Giemsa stained blood smears. Result indicated that seasonal infection is a character of this disease and Arthropods is a key to it. Infection rate increased from cold winter to warm and damp spring. 80% sheep can be infected by Eperythrozoon ovis in hot summer generaly.Two primers were synthesized based on the conservative sequence of Eperythrozoon ovis 16S rRNA. A 1000bp fragment was amplified by using polymerase chain reaction. The amplified fragments with the expected size were identified by EcoRI restriction digestion.The crossing-reaction and specific-reaction indicated that PCR is quick ,sensitive and special.lt is a effective way to be used to diagnose Eperythrozoon ovis from the whole level.The optimal condition of ELISA was established in this experiment. The dilution of antigen positive serum and HRP-goat anti rabbit IgG is 1:64,1:160 and 1:1000 respectively. The optimal reaction time of antibody and HRP-goat anti rabbit IgG is Ih.The minimum detection of Eperythrozoon ovis is 50.30ug/ml.The crossing-reaction indicated that ELISA is a quick, sensitive way adapt to colony check.
