Facters affecting formation of somaclones of Aloe were studied at first. And then callus was induced to produce variant by mutagen EMS. The variant was selected by selective pressure HYP and it could resist low temperature. And the relative biochemical and physiologic characteristic of cold-resisting of the HYP-resistant variant ,its regenerated plantlet and the callus derived from the regenerated plantlet were studied. The results showed that: The best explant is stem for callus inducing of Aloe. And frequency of callus inducing and differentiation of A.aisis.Mill is higher than that of A.veraL.var.chinesis(Haw)Berg. and A.arborescens.Mill. Additive GIn is good to improve inducing frequency. Lower concentration of HYP is good to improve frequency of differentiation. The best inducing medium is MS add NAA 1 .Omg/L,6-BAO.O2mIL,Gln5mg/L,the best differentiation medium is MS add NAAO.Smg/L, 6-BAO.2mg/L,HYP2.O-2.5mmol/L.The medium for root formation is l/2MS add NAAO.Smg/L,IBAO.2mg/L. Time of treatment and concentration of EMS affect the frequency of differentiation of callus and plantlet. The proper combination of concentration of EMS is 0.5% and 3 hours. The half death concentration of HYP is 13.Ommol/L and large scale death concentration is 20.Ommol/L. HYP-resistant variation and its regenerated plantlet and the callus derived from regenerated plantlet remain characteristic of high proline, osmotic stress-resisting and cold- resisting. Meanwhile the regenerated plantlet has variation of POD iosenzyme.
