Aloe is a perennial berbaceous plant which belongs to the lily family. It grows mainly in the dry regions of Africa. Its thick leaves are rich in pulp that contains many kinds of active matter. Modem investigation testified that aloe has the efficacy of purgation, quench evil heat in the liver, let off heat, resolve bladder, destroying parasites, diminish inflammation, anti-ultraviolet radiation, antivirus, raise immunity, enrich hair and etc. Now, the aloe has been widely applied to the fields of medicament, foodstuff, cosmetic and others. Aloe is in great demand with the development of the aloe industry, traditional methods for aloe propagate are cuttage and root division. These methods are limited by the propagation radix. They also easily induce the virus accumulation, influence the grow and even make the plant degenerate. Now, the asexual propagation technique has been an important method for aloe propagate. In 1974, Groenewad E successful planted an aloe heal organization plant from Pretoria aloe seed. From then on, the investigation into aloe high frequency propagation go far continuously in the world. Now, the relatively integrated asexual high frequency propagation system has been founded. In recent years, more and more scientists begin to deal with the investigation into aloe transformation and have obtained some progeny.In this article, in order to obtain a new breed of aloe with function of dispel scar, we transfer bFGF into aloe stem. We hope our work can offer a new material for cosmetic, so that aloe can contributes to people more. We draw the following conclusions.1. Aloe genetic transformation system have been set up, explored effective methods for deferent aloe to induce buds, subculture and root propagation, which have been considered as the base for aloe transform. As a result, the best sterilization condition is that disinfected for 30 seconds in water with 70 percents of alcohol firstly, and then disinfected for 8 minutes in water with 0.1 percents of mercuric chloride. Buds can be induced from two kinds of the aloe, however, the rate of inducing is up to genotype, deference medium, concentration of hormone. For deferent typies aloe inducing, the inductivity of improved MS medium which has the highest rate of inducing buds is better than 1/2MS medium, and the worst are B5 and N6 medium. But for hormone, when its concentration is 6-BA2mg/L+NAA0.15mg/L, all kinds of aloes have a high inductivity. MS+6-BA4.0mg/L+NAA0.1mg/L is the best medium for subculture and MS+NAA2.0mg/L with activated carbon in 0.3%is the best medium for root propagation.2. Genetic transformation system of aloe mediated by Agrobacterium trmefaciens: By light of Agrobacterium tumefaciens, translate bFGF gene to aloe arborescens Mill., investigate the conditions, and discuss the problems just like deferent kinds of bacterial strains, deferent concentrations of infestations, deferent times for infesting. Use the sturdy individual aloe stem basal part as the explant. 3~5 days preculture of the explants before Agrobacterium tumefaciens had a positive effect on infected by the transformation. The bacterium density was OD600 0.6~0.8, EHA105 was the best bacterial strain and the infection time was 25 minutes. Explant was cultured in MS+sucrose10g/L+100μmo/LAS for 3 days, selected in MS+6-BA3mg/L+Cef 400mg/L+Kan50mg/L for 60 days, differentiated in MS+6-BA2mg/L+NAA0.15mg/L, and transplanted after the buds rooted. By the number of seedlings, transformation percentage is 4.8%.It can be tested by PCR and Southern that when the buds were differentiated and developed plant, which indicated that bFGF gene have been transferred into aloe successfully.
