The Agrobacterium Mediated Transformation and Expression of fl-i, 3-Glucanase Gene in Cucumis melo L. Li Qun(Plant Physiology & Biochemistry) Supervised by Prof.Li Guan Abstract In order to enhance the fungal resistance of Xinjiang muskmelon ,the research for introducing ~8 -1 , 3-glucanase gene into muskmelon mediated by Agrobacterium tumefaciens was done. Using the cotyledon pieces of muskmelon as explants, a regeneration protocol for Xinjiang muskmelon have been established,and this protocol was combined with Agrobacterium tumefaciens transformation to produce transgenic muskmelon plants. Cotyledon explants were cocultivated with A.tumefaciens carrying a Ti plasmid in which a fl -1, 3-glucanase coding sequence as well as the neomycin phosphotransferase LI(NPT U)coding sequnence were fused to the Cauliflower Mosaic Virus( CaMV )35S promoter.Under optimal condition,the transgenic muskmelon plants were obtained and were normal in morphology. PCR specific gene arnplification,SDS-PAGE,and peroxidase isoenzyme PAGE were used to detect the integration and expression of the purpose genes.The results indicated that the foreign gene did integrate within the genome of the regenerated plants and was expressed normally.
