Development Of Isozyme And RAPD Makers Of Phytophthora Sojae

Four isozymes of 15 isolates of? sojae and 8 isolates of seven other Phytophthora species were studied using polyacrylamicide gel electrophoresis. On the other hand, DNA of these 23 isolates was amplified with 12 random primers. The results were summarized as follows: Significant differences of EST and SOD zymograms among P sojae and seven other Phytophthora species indicated that EST and SOD isozyme zymograms were useful and reliable supplementary tools for identification of? sojae. There were no marked differences of DIA and CAT zymograms among? sojae and seven other Phytophthora species~ so they can抰 be used to identify P sojae. SOD, CAT and DIA isozyme zymograms of? sojae were first reported in this study. 152 RAPD markers were obtained, 842% were polymorphisms. A dendrogram depicting the relationship among 23 isolates of Phytophthora was constructed by hierarchical cluster method. The genetic diversity among Phytophthora species was detected, and? sojae was far away from seven other Phytophthora species in genetic distances. Moreover, the special RAPD markers of? sojae were found. The results showed higher similarities among 15 isolates from? sojae in four isozyme zymograms, and there was no interrelation between DNA polymorphism of isolates of? sojae and theirs geographical origins and virulence patterns. RAPD analysis also indicated that there existed the genetic diversity Within? sojae isolates, and there was no interrelation between DNA polymorphism of isolates of? sojae and their geographical origins, but the relationship between virulence patterns and RAPD markers was presented in some isolates.
