Abstract/Summary:
Phytophthora soja is an important plant pathogenic oomycete,and the soybean disease caused by it has great harm to soybean production and can cause huge economic loss.Previous studies have shown that the pathogenicity of Phytophthora sojae can be divided into different pathogenicity types or races,and the relationship between the soluble protein and esterase isozyme of Phytophthora sojae and the relationship between the pathogenicity types or races of Phytophthora sojae is little known.In this thesis,34 strains of Phytophthora sojae in Anhui province were studied by SDS-polyacrylamide gel electrophoresis(SDS-PAGE)and PAGE electrophoresis,and their genetic differences were studied at the level of protein and enzymology,and a dendrogram was constructed to determine the electrophoresis patterns of soluble protein and esterase isozyme of different strains.The correlation between the difference of electrophoresis patterns and the geographical origin and pathogenicity of the strains was discussed.The possibility of classification and identification of Phytophthora sojae by electrophoresis analysis of soluble protein and esterase isozyme was discussed,in order to provide the experimental ecidences for the breeding and rational use of soybean varieties resistant to soybean blight and for the integrated management of the disease.The main results are as follows:1.SDS-polyacrylamide gel electrophoresis(SDS-PAGE)was performed on 34 strains of Phytophthora sojae of different sources in Anhui province.,respectively,to isolate the article 9 ~ 15 visible band,through calculation and analysis of soybean phytophthora Rf value of 0.08 ~ 0.88,the characteristic band of Rf value of 0.16,0.28,0.36,0,1,0.6,0.68 and 0.6,a total of seven.The soluble protein map was associated with geographic origin.2.PAGE electrophoresis was carried out for 34 strains of Phytophthora sojae from different sources in Anhui province.All the tested strains had 1 clear bands with Rf value of 0.59.The difference between bands was shown on the width and color of the strip.The cluster analysis showed that the similarity of the esterase Isozyme of the strain was independent of the geographical origin.3.The correlation analysis of the soluble protein atlas of the tested strains and the esterase isozyme patterns and pathogenicity showed that the soluble protein atlas of the strain and the esterase isozyme atlas had no significant correlation with the pathogenic type,but all of the tested strains had 1 clear bands of 0.59 Rf value,which could be used as reference index for identification of Phytophthora.
