In the present studies.the Aelnv antisense gene was transformed by the Agrobacteriun2 turnefaciens, LBA4404 in which the plasmid pBIRA is the vector for the Gene expression.Also.the optimum transformation system and the regeneration system of leaf discs and stem segments were studied.the gene was cloned from potato tuber. which stored in 4C.the results showed that: In the optimized regeneration s stem.the callus induction media for the two kinds of explants were:Stem segments:MS+NAA 0.2mg/L?BA ~ 25mg/L+GA3I.Smg/L, leaf of didcs:NAA 0.2mg/L-r6BA 3.4mg/L+GA-, 3.Omg/L. The regeneration media for Stem segments was:MS+6BA3.4mg/L+GA~ 6.Omg/L. The selected media for rooting was: MS+NAA 0.lmg/L. The selective pressure during the callus induction and regeneration was Kanamvcin I OOmg. In the optimum regeneration system.the optimum bacterium concentration in value of 0D600 was 0.5, it had a best promoting effect on stemsegments and the leafier discs.the optimul time of preculture,infection and co-culture for leaf discs were 3 days. I 2mins and 3 days respectively; to stem segments.they were 2 davs.8mins and 2 days. Nine cultivar were chosen for transformation. In the transformation of potato using Aclnv Antisense gene .transgenic plants were obtained from the callus derived from the stem segments of tlanticultivar.Using PCR methods assay showed that the regenerated plants had postive reaction, thus it could be proved that they are transfonned plants, the primary detection expressed that the Aclnv antisense gene have transformed into potato genome...
