Potato is cultivated and studied extensively, whatever as one of important economical crops, which supplies edible starch for human, raw material for pharmaceutical industry and other related region, or as one of model plants, which provides abundant genetic information for scientific research. In traditional flora of plant, potato is located in Solarium under Solanaceae and is described as annual herb with underground tuber and possess authentic name "Solanum tuberosum". Due to abundant starch in tuber, the wild potato is introduced as food crop from its original place of the South American and, then of cultivated environment difference and unintermittent breeding, many potato cultivar were produced. Whereas many pathogen including fungi, virus and bacteria can attack or infect potato tuber, stem or leaf within any phase of its growth period, and many disease including late blight, soft root and leaf roll etc by these pathogen usually lead to devastating reduction of production of tuber and even nothing, such as the outbreak of late blight which caused by the oomycete plant pathogen Phytophthora infestans lead to the Irish potato famine in 1854. In addition to, many insects can hinder potato's growth by gorging its leaf, suck its sap and eventually impair development of tuber. So, many measures were taken to resolve or control the pests and pathogen.Chemical control by spray lethal pesticide to kill pests is used widespread, but the pests were killed and many beneficial insects also were put to death and predatory birds or some parasitoid in those pests were impair seriously. Not so that, abuse of pesticide have been lead to a disastrous cycle that many pests have been formed resistance to pesticide and so high-dosed pesticide be required to use or new strong toxic pesticide be forced to produce, in turn the pests will evolve to stronger resistance before so strong powerful select pressure and no enough pesticide to be used in no far the future.Biological control is a good measure to protect crops from hurt of pests by interaction between species through food webs. However, the pure biological control to pests mainly dependent onpredatory number and need long time to attain expected result, and not meet the rapid development of modern agriculture which needs effective methods to control pests to minimize the damage by pests.Combinating advantage of biological control and toxic character of pesticide in chemical control, the genetic engineering offers immense opportunites to control pests by incorporate resistance gene coding toxin or chemical matter to interfere growth or development of pests into potato. The snowdrop lectin(GNA) was isolated from bulbs of Galanthus navil of Amaryllidaceae showed averse effect on insects, especially suck-saping insects such as aphids, planthopper and have been incorporated into crops plants including rice tobacco and potato etc by Agrobactium tumefaciens mediated to improve their resistance to insects. On potato genetic transformation with GNA, many study have been performed by foreign researcher and not be found in China. So the potato plants cultivated in SiChuan province were selected to study genetic transformation with GNA and expect to improve their character to resistant to pest. Many genetic transformation technology were taken to transform potato and gain satisfied results. Agrobacterium tumefaciens mediated transformation method was performed widely to transfer foreign DNA in experimental plant and be proved successful to transform potato. However the foreign DNA not be transfer directly into potato by A.tumefaciens because A.tumefaciens can cause tumor by interfering the normal synthesize of genetic information of host plant genome. In general protocol, foreign DNA was integrated firstly with expression vector and then be delivered into A.tumefaciens and into potato plant in the end by A.tumefaciens mediated. Many expression vectors can be used to perform the task and the expression vector pCAMBIA-lecGAN2 was constructed and is used in our experime...
