A series of experiment had carried out on the temporal/spatial expressionof B.t transgene cotton, the variation of enzyme in bollworm which werefeeded on the leaves of B.t transgene cotton and chemical regulation of B.ttransgene cotton expression.1.All kinds of tissues from B.t transgene cotton have good ability to kill thelarva, such as Growth Survive a~d Development. The larva which feed on B.ttransgene cotton had lower survival rate, lighter weighty and shorterdevelopment time than control larva. B.t protein in the same tissue can beexpressed at diffei'ent level at different time and B.t protein in the differenttissue can also be expressed at different level at the same time. During thelatter life of 13.t transgene cotton the amount of E3.t protein isn't enough tocontrol bollworm efficiently, so the B.t transgene cotton need pestmanagement, it is that chemical pesticides should be used to kill bollworm orsome other methods should be applied to assure the cotton production.2.Through bioassay and biochemistry methods, I researched the dynamics ofa-naphthylaeetate esterase and acetylcholine esterase (ACE) in the resistantpopulation. The result proved the ACE of the bollworm which feed on theleaves of B.t transgene cotton is more active than the ACE of the controlbollworm, but the dynamics of a-naphthylacetate esterase doesn't changeregularly. By bioassay, the chemical pesticides resistant bollworm is moresusceptive than the control bollworm to ~.t transgene cotton. The ACE of thesurviva! bollworrn feeded on B.t cotton is more active, so it indicated that thedynamics of ACE is an important factor to bollworm which get resistantability to B.t pesticide.3.Salicylicc acid can regulate many genes in plants, but in C-i transgene cotton,the promoter of gene is 35S, w~iich can't be regulated by SA, so SA can'tincrease the B.t gene expression. Shericca Williams use pr-la promoter to B.tgene and get the B.t transgene cotton which can be regulate by SA. These results provide the novel way to construct transgene plants.
