Abstract:To obtain ehalkbrood disease correlative gene fragment of west honey bee. The random amplified polymorphic DNA(RAPD)analysis was made in susceptible and resistant genome of west honey bee with 16 random primers. Only one out of nine primers indicated the polymorphic DNA between susceptible and resistant west honey bee. The polymorphic bands were assigned to be P14330.Using the polymorphic primer 14 to detect two breeds of west honey bee susceptible to chalkbrood disease and three breeds of those resistant to chalkbrood disease. RAPD showed the same results more than ten replications. P14380 was only amplified in susceptible west honey bee and absent in all of the resistant west honey bee. Further 380bpdigoxigenin-labeled probe was used to detect Southern blotting of RAPD productions and dot blotting of genome DNA. Two hybridizations spot or band was only appeared in two susceptible breeds but absent in three resistant breeds. 380bp fragments was also sequenced. These observations indicate (1) P14380 is a special gene marker of west honey bee susceptible to chalkbrood disease.(2) 3 8Obpdig-labeled probe can be used to screen susceptible and resistant west honey bee.(3) 380bp bands tightly correlated to chalkbrood disease gene of west honey bee. Probably the susceptible gene which was inserted increased the primer-binding point, then the special DNA band was amplified. The P 14380 markers obtain by the method of RAPD not only provide a new short cut to detect west honey bee susceptible toIIchalkbrood disease, but also has instructive meaning using transform gene technology for improving honey bee breeds.Post-graducate GuoXiaoHongDirected by Pro.
