The Genetic Analysis And Molecular Mapping Of Euil Gene In Rice

XQZ eB(l) (XieQingZao eB(1)),an elongated-uppermost-internode mutant,was obtained from maintainer line XQZ B(XieQingZao B) by irradiation. Compared with the origin XQZ B, the plant height and length of the uppermost internode, second and third internode from top of XQZ eB(l) were significantly increased. The plant height and culm type of F1, F2 and B1F1 of the crosses XQZ eB(1)/XQZ B and XQZ eB(1)/ AJNT(AiJiaoNanle) as well as parents were studied. The semidwarf culm type of F1 was similar to the semidwarf parent XQZ B or AJNT. The population of F2 and B1F1 was devided into two different groups; one was the short culm group, the other was long culni group with elongated uppermost internode. The X 2 test of the ratios of long culm to short culni in F2 and B1F1 was conformed to 3:1 and 1:1, respectively. The results above indicated that the long culm character of XQZ eB(l) was governed by a single recessive gene .The allelisim test demonstrated that the mutant gene of XQZ eB(l) was allelic with euil of IR5Oeui (derived from 76:4512). SSR maker is a new molecular maker based on PCR technique. A F.1 population was developed from the cross of XQZ eB(l)/ AJNT to identify the maker linked to the euil. One maker RM164 linked to euil gene was screened by combining SSR molecular maker with BSA (bulked segregation analysis) method. The euil gene was mapped on the long arm of 5 chromosome. The genetic distance of RM164 to euil was 18.4cM by Mapmaker/EXP3.O analysis. The maker linked to euil gene may facilitate the establishment of MAS(maker-assistant selected) method for eui-hybrid breeding system and build a foundation for fine-mapping and cloning of eui] gene.
