Given condition, the regenerated tobacco K326 plantlets, which were produced from the leaves of the sterile seedling of tobacco, were treated with 0.3mg/L 2,4-D and mutant of fast-growing stain in R. sp.vigna. Many para-nodules similar to sphere formed from their roots. The para-nodule was composed of epidermis cells, cortex cells and meristematic cells. The major part of the para-nodule base which joined up the tobacco root were parenchyma cells, their intercellular spaces were obvious. The studies of electron microscope indicated that there were a lot of rhizobia in the intercellular spaces and a few rhizobia in the cells. Usually these rhizobia hadn't peribacteriod membranes, but they were sometimes division.The para-nodule was obvious different from the fake root. The later was cone, its top was root cap following meristematic cells, the elongation cells joined up the root, and there weren't rhizobia. The fake root may be induced by 2,4-D because 2,4-D can change the arrange of microtubule.There were many the protein bodies (PB) in the tobacco para-nodule. Some were similar to sphere or oval, others were irregular. The main distribution region of the PB was the vacuole. Their electron density was high, sometimes there were some vesicules in the PB. There weren't obvious membrane surrounding their surface. The PB's origin may be the protein's deposition in the vacuole. And we studied the PB's development in detail.ATPase were located at the para-nodule's cell wall, plasma membrane and some also existed in plasmodesmata, nucleus, mitochondria, endophasmic reticulum and rhizobia. Besides the cell's type and development level, ATPase activity was related to the region of requiring energy. ATPase can let ATP divide ADP and ~P, supplying sufficient energy for the cell's growth, development, the substance transport and nitrogen-fixing.ACPase was a kind of hydrolytic enzyme. Its distribution and activity were directly related to the cell's development. In the young cells, ACPase existed in the para-nodule's cell wall, plasma membrane, plasmodesmata, cytoplasm, mitochondria, endophasmic reticulum, protein body etc. In the process of cell senility, ACPase activity was strengthened in cell wall, plasma membrane and organelle etc. In the former, ACPase may be related to cell wall's metabolism, the substance transport and the phosphorus matter metabolism during the PB development. In the latter, it may be related to the cell's disintegration.
