Because the resource of local breeds in China alfalfa is disarrangement , genetic diversity of eight samples in Medicago sativa were studied using SSR technique. Genetic heterozygosity, Polymorphism information content, Similarity and Genetic distance was calculated among eight samples,eight samples were clustered using three methods.Their relationship was inspected, The main results were summarized as the folowmgs: 1 .Amplification with 6 pair microsatellite primers detected 54 amplified fragments The polymorphic fragment was 51 bands ,communal fragment was 3 bands.The amplified fragment with different primer was between 5-17, average was 9 and the proportion of polymorphic fragment was 94. 44%. SSR can detect polimorphism sensitively,so most of the microsatellites used in this research can be used as marking of linkage analysis.2. genetic consistency of Huanghou alfalfa , Zhunge'er alfalfa , Caoyuan No, 2 alfalfa , Heping alfalfa , Bailv alfalfa, Zhaodong alfalfa, Aohan alfalfa, Gongnong No.l alfalfa was small,and most of them were heterozygous locus,and hereditary variation was greate.3. hereditary variation of local breeds in China alfalfa was chief from each samples,and local breeds in China alfalfa was short of specific property and have narrow genetic background. 4.The scope of genetic distance was 0.162-0.503 among eight samples.The variation was biggest between Gongnong No.l alflalfa and Heping alfalfa and was lowest between Bailv alfalfa and Zhaodong alfalfa.At 0.25of genetic distance,eight alfalfa examples were classified into four groups: the first group include Huanghou alfalfa ,Zhunge'er alfalfa and Caoyuan No.2 alfalfa;the second include Heping alfalfa;the third include Bailv alfalfa and Zhaodong alfalfa;the fourth include Aohan alfalfa and Gongnong No. 1 alfalfa.
