The experiments of somatic nuclear transfer were conducted by use of bovine ear fibroblast as donor and rabbit oocyte as recipient. The results showed that 202 reconstructed eggs were obtained by the fusion of five kinds of bovine ear fibroblasts (fresh or refrigerated non-Go cells,fresh or refrigerated cells at 100% cofluency and refrigerated serum-starved cells) and 436 rabbit oocytes,the fusion rate was 46.3%,and in 180 reconstructed embryos cultured in vitro,112 of them developed to 2-cell(62.2%),26 developed to morula (14.4%),20 developed to blastocyst (11.1%),which suggesting that bovine somatic cells could be dedifferentiated in rabbit oocytes,and reconstructed embryos in inter-species could develop to hatched blastocyst. The findings of this research also indicated that bovine ear fibroblasts preserved at 4 within 19d or frozen at -196 had the normal proliferating and wall-attaching ability when they were re-cultured;the cells refrigerated at 4 for 14d could still serve as donor;mortality rate of reconstructed eggs were increased significantly when adding cytochalasin (CB) into fusion solution (P<0.05);there's no significant difference in fusion rate of reconstructed eggs when using oocytes from multiparous or young rabbits as recipients (P>0.05),but the mortality rate of eggs reconstructed with multiparous rabbit oocytes was significantly higher than those with young ones (P<0.01);the embryos reconstructed with serum-starved cells were significant higher in the rate of 8-16-cell or morula stage (PO.05),and blastocyst stage (PO.Ol)than those with non-Go cells.
