Studies On The Karyotype And Banding Of Chromosome Of Northeastern Wild Boar And Its Hybrid With Swine

In this experment, the karyotype, C-banding, G-banding and high resolution G-banding of chromosomes of Northeastern wild boar and its Hybrid with swine were studied. We found the fittest conditions of lymphocyte culture, and received Karyotype. By using usual method, we got C-banding, and approximate 290 bands level of G-banding of chromosomes, and by using micro-Colchicin method, we also obtained approximate 410 bands level of High resolution G-banding of chromosomes. This study was to elementarily affirm taxonomic status of Northeastern wild boar, and to provide basic data of Karyotype and banding of chromosomes for studying genesis, evolution and breed acquirement of swine and gene localization of boar.The results were as follows:1 . The fittest PH of culture medium of lymphocyte culture of Northeastern wild boar is 7.2-7.4, and that of the Hybrid is 7.0~7.2; the suited consistency of Colchicin is 0.05ug/ml toward lymphocytes of Northeastern wild boar and its Hybrid with swine, and the functional time of Colchicin is 3h; the suited hypotonic time is 20 min under the condition of KC1 hypotonic fluid of 37"C, 0.075mol/L.2. By adding to Glutamine, we may use micro-Colchicin method and Trypsase-EDTA method to get perfect high resolution G-banding of Northeastern wild boar and its Hybrid with swine.3. The chromosome numbers of Northeastern wild boar and its Hybrid with swine are same that of swine, all are 2n=28. Their karyotypes have no obvious difference from those of swine.4. The C-banding of chromosomes of Northeastern wild boar and its Hybrid with swine has polymorphism, the numbers of C-banding of the hybrid are more than that of Northeastern wide boar.5. The most bands of G-banding, of high resolution G-banding of Northeastern wild boar and its hybrid with swine and of swine are same; in the different bands, the bands of the hybrid and of Northeastern wild boar are mostly similar, but comparing with them, the bands of swine are less similar; the differences of G-bands of the three kinds lies in the location, breadth and whether specific bands of specific chromosomes exist or not, and the most differences will appear on the high resolution G-banding.
