Locusta migratoria manilensis (Meyen) is subordinated to Orthoptera,Acridoidea,Oedipodidae ,Locusta Linnaeus . It is a major pest in agriculture, distributed the Europe, Asia, Africa and America. According to the study of Locusta migratoria manilensis (Meyen), it contains locust protein, fatty acid, lecithoid, SOD, flavone mine and so on. Locusta migratoria manilensis has been developed into products. There are a lot of research works on morphologic, histology, procreation, migration of Locusta migratoria manilensis (Meyen), but little about nucleus gene and chromosome fields.On the cell level, the major hereditary material exists in chromosome. Organism character is controlled by gene in chromosome. When structure or quantity of chromosome changes, organism karyotype changes, which will influence it's heredity, variation, evolution. Therefore, by analyzing and researching on chromosome, we are able to know the origination, evolution, the law of heredity and variation of organism, to judge the relationships between organisms. The research on chromosome of Locusta migratoria manilensis is not much in China. Mostly on biochemistry, physiology etc. To exploit and utilize Locusta migratoria manilensis better, avoid disastering the farming yielding, we carried out preliminary research for the chromosomes of Locusta migratoria manilensis.In this paper, spermary cell chromosome specimen of Locusta migratoria manilensis was prepared through injecting colchicine (0.04%, 0.1ml each) to living locust, and made the spermary cell suspension, then dripped the suspension on slide. It is studied the karyotyp, C-banding, Ag-NORs and G-banding. The chromosome of Locusta migratoria manilensis is Conventional chromosome, amount is 2n(♂)=22+X0=23.The biggest chromosome is X chromosome. The all chromosome is end centromere chromosome, NF=23. C-banding displays on the centromere, and all chromosomes are end centromere chromosome. The number 5, 6, 11 chromosome centromere colorated lighter. Number 1,2,3,7,8,9,10 chromosome centromere colorated dark, number 4 and X chromosome centromere colorated . And number 2, 3 chromosome have mediacy banding. The Ag-NORs orientated on the number 2 chromosome. And all genome has 134 pieces G-banding.To afford data for identifying the relative of insect, community frame and resource of insect, we pilot study the Locusta migratoria manilensis chromosome. It is set base on orientation of AFLP and fluorescence in situ hybridization, FISH.
